Role of orexin-1 receptors in the dorsal hippocampus (CA1 region) in expression and extinction of the morphine-induced conditioned place preference in the rats

Orexinergic system is involved in reward processing and drug addiction. Objectives here, we investigated the effect of intra-hippocampal CA1 administration of orexin-1 receptor (OX1r) antagonist on the expression, and extinction of morphine-induced place preference in rats. Conditioned place preference (CPP) was induced by subcutaneous injection of morphine (5 mg/kg) during a 3-day conditioning phase. Two experimental plots were designed; SB334867 as a selective OX1r antagonist was dissolved in 12% DMSO, prepared in solutions with different concentrations (3, 30, and 300 nM), and microinjected into the CA1 and some neighboring regions (0.5 μl/side), bilaterally. CPP score and locomotor activity were recorded during the CPP test. Results demonstrated that intra-CA1 administration of the OX1r antagonist attenuates the expression of morphine-induced CPP. Furthermore, higher concentrations of SB334867 facilitated the extinction period of morphine-induced CPP and reduced its latency. Nevertheless, solely administration of DMSO did not have any influence on the CPP scores and locomotion in both phases. Our findings suggest that OX1rs in the CA1 region of the hippocampus are involved in the expression of morphine CPP. Moreover, blockade of OX1rs could facilitate extinction and may extinguish the ability of drug-related cues. It seems that the antagonist might be considered as a propitious therapeutic agent in suppressing drug-seeking behaviors.

[D-Leu-4]-OB3 and MA-[D-Leu-4]-OB3, small molecule synthetic peptide leptin mimetics, improve glycemic control in diet-induced obese (DIO) mice

We have previously shown that following oral delivery in dodecyl maltoside (DDM), [D-Leu-4]-OB3 and its myristic acid conjugate, MA-[D-Leu-4]-OB3, improved energy balance and glucose homeostasis in genetically obese/diabetic mouse models. More recently, we have provided immunohistochemical evidence indicating that these synthetic peptide leptin mimetics cross the blood-brain barrier and concentrate in the area of the arcuate nucleus of the hypothalamus in normal C57BL/6J and Swiss Webster mice, in genetically obese ob/ob mice, and in diet-induced obese (DIO) mice. In the present study, we describe the effects of oral delivery of [D-Leu-4]-OB3 and MA-[D-Leu-4]-OB3 on glycemic control in diet-induced (DIO) mice, a non-genetic rodent model of obesity and its associated insulin resistance, which more closely recapitulates common obesity and diabetes in humans. Male C57BL/6J and DIO mice, 17, 20, and 28 weeks of age, were maintained on a low-fat or high-fat diet and given vehicle (DDM) alone or [D-Leu-4]-OB3 or MA-[D-Leu-4]-OB3 in DDM by oral gavage for 12 or 14 days. Body weight gain, food and water intake, fasting blood glucose, oral glucose tolerance, and serum insulin levels were measured. Our data indicate that (1) [D-Leu-4]-OB3 and MA-[D-Leu-4]-OB3 restore glucose tolerance in male DIO mice maintained on a high-fat diet to levels comparable to those of non-obese C57BL/6J wild-type mice of the same age and sex maintained on a low-fat diet; and (2) the influence of [D-Leu-4]-OB3 and MA-[D-Leu-4]-OB3 on glycemic control appears to be independent of their effects on energy balance. These results suggest that [D-Leu-4]-OB3 and/or MA-[D-Leu-4]-OB3 may have application to the management of the majority of cases of common obesity in humans, a state characterized at least in part, by leptin resistance resulting from a defect in leptin transport across the blood-brain barrier. They further suggest that these small molecule synthetic peptide leptin mimetics, through their influence on glycemic control, may prevent the pre-diabetic state associated with most cases of common obesity from escalating into overt type 2 diabetes mellitus.

Members of the Bacillaceae family, including Bacillus spp., Brevibacillus spp., Paenibacillus spp., Aneurinibacillus sp., and Halobacillus sp., are an important source of structurally diverse classes of short peptides of ∼ 30 residues or fewer possessing peculiar and rapid killing activity against various pathogens. Additionally, many have unique structures that enhance resistance to hydrolysis by proteases, and these are ideal therapeutic tools and potential alternatives to current antibiotics. The need for novel antibiotic lead compounds is urgent, and this review summarises 119 Bacillaceae compounds published since 2000, including 12 surfactin-like lipopeptides, 16 iturinic lipopeptides, fengycin C, 33 other cyclic lipopeptides, 26 linear lipopeptides, two thiopeptides, four 2,5-diketopiperazines, 20 typical cyclic peptides, and five standard linear peptides. The current and potential therapeutic applications of these peptides, including structure, antibacterial, antifungal, and antiviral activities, are discussed.

Cloning and gene expression analysis of two cDNA of cysteine proteinase genes involved in programmed cell death in the inner integument from developing seeds of Jatropha curcas L

In this paper, two cysteine proteinases were cloned from Jatropha curcas seeds. The full length cDNAs obtained from cloning of Jc-CysEP1 and Jc-CysEP2 genes were 1.516bp and 1500 pb, respectively. The Jc-CysEP1 contained a 1083bp open reading frame (ORF) coding for 360 amino acids. The JcCysEP1 protein sequence had an estimated native molecular weight of 36.89 kDa, with a predicted isoelectric point of 4.55. The average lengths of JcCysEP1 5′ UTR and 3′ UTR were 269 bp and 167bp, respectively. The Jc-CysEP2 contained a 1077 pb open reading frame (ORF) that encoded 358 amino acids. We also identified UTRs with lengths of 229 pb (5′UTR) and 194 pb (3′UTR). The Jc-CysEP2 sequence had a native molecular weight of 39.94 kDa, with a predicted isoelectric point of 6.19. Real-time PCR analyses of developing seeds (stages I-VII) showed that most cysteine proteinase genes were expressed at stage IV (middle stage) revealing peculiar spatio-temporal differences. JcCysEP2 was the cysteine proteinase gene with the highest expression in inner integument tissue, while JcCysEP1 was expressed in lower levels. Our results suggest that JcCysEP2 could be the major cysteine proteinase gene involved in PCD events in inner integument tissue, playing a critical role in PCD events during seed development, while Jc-CyEP1 and JcCysEP2 genes act cooperatively in stages IV-VII. JcCysEP2 is important to complete their participation in PCD until development of seeds.

Cardiovascular monitoring of cognitive workload: Exploring the role of individuals’ working memory capacity

Cardiovascular measures have been found to be sensitive to task onset and offset, but are less sensitive to adjacent levels of increasing cognitive workload. A potential confound in the literature is the disregard of individual differences in cardiovascular reactivity. In particular, the individuals' working memory capacity (WMC) is likely to play a role in cardiovascular reactivity to workload. A total of 98 university students performed four cognitive tasks that varied in their level of workload. The operation span (OSPAN) task was used to measure the participants' WMC. A variety of cardiovascular measures were gathered in real time during the experiment. Derived measures of blood pressure regulation were also calculated. In line with what was hypothesized, cardiovascular measures detected workload onset and offset but did not consistently distinguish between the individual task levels. Furthermore, a significant interaction between workload levels and WMC showed that the individuals' cardiovascular profile varied depending on their WMC scores. In addition, WMC negatively predicted subjective ratings of task difficulty as well as task performance, with subjective estimation of task difficulty and error increasing as WMC decreased. The results suggest that WMC may play a critical role in determining how individuals react to increased cognitive workload.

Four-weeks versus 5-weeks hypofractionated high dose radiotherapy as primary therapy for prostate cancer: interim safety analysis of a randomized phase 3 trial

PurposeHypofractionated radiotherapy (HFRT) for localised prostate cancer (PC) is safe and effective. The question remains which hypofractionation schedule to implement.In this study 2 different HFRT regimens are compared.Methods and MaterialsBetween 6/2013 and 7/2016, 160 PC patients were randomly assigned (1:1) within this single centre phase 3 trial, to 56 Gray (Gy) (16x3.5Gy, Arm A) or 67 Gy (25x2.68Gy, Arm B). Randomization was by computer-generated permuted blocks, stratified on prior transurethral resection of the prostate and presence of a dominant intraprostatic lesion. Treatment allocation was not masked. Clinicians were not blinded. The primary endpoint is acute gastro-intestinal (GI) toxicity, assessed by CTC version 4.0 and RTOG. An interim analysis on acute toxicity was planned on 160 patients to prove the safety of both treatment regimens. If ≥22/72 patients have grade ≥2 GI toxicity, the study arm is to be rejected. The study is registered on Clinicaltrials.gov (NCT01921803).ResultsIn Arm A, 20 (26%) and 1 (1%) patients developed acute grade 2 and grade 3 GI toxicity. In Arm B, 16 (20%) reported acute grade 2 GI toxicity.resultsIn Arm A, 42 (55%) and 5 (6%) patients developed acute grade 2 and grade 3 urinary toxicity. In Arm B, 40 (49%) and 7 (9%) reported acute grade 2 and grade 3 urinary toxicity.Toxicity peaked during radiotherapy resolving in the months following radiotherapy.ConclusionWith acute grade ≥2 GI toxicity reported in 21/77 patients in Arm A and 16/82 patients in Arm B, both treatment arms are considered safe.conclusion

Prospect of robotic assistance for fully automated brachytherapy seed placement into skull base: Experimental validation in phantom and cadaver

Background and purposeTo investigate the feasibility and accuracy of robot-assisted brachytherapy for skull base tumours.Material and methodsA custom robot system was tested on both phantom and cadaveric specimen. Cone beam CT (CBCT) images were transferred to the graphical user interface (GUI) for planning trajectories and the data were sent to the robot control unit. Following registration, the puncture needle was inserted into the target by the robot under navigation guidance, and seeds were implanted. Placement error was instantly displayed on the GUI; the result was verified after postoperative image scanning.ResultsA total of 150 seeds (100 for phantom experiments, 50 for cadaveric studies) were deposited by the robot system. In phantom experiments the mean placement error was 0.57 ± 0.21 mm (measured by the navigation system) vs. 1.41 ± 0.38 mm (measured by image fusion) (p < 0.001); in cadaveric studies the corresponding figures were 0.60 ± 0.30 mm vs. 2.48 ± 0.32 mm (p < 0.001). There was no significant difference for comparison of accuracy test in phantom experiments (p = 0.173) as well as in cadaveric studies (p = 0.354). Accuracy was better in the phantom experiment than in cadaveric studies (p < 0.001).ConclusionsThe performance of robot-assisted skull base brachytherapy is feasible and accurate. Dosimetric coverage will need to be demonstrated in further studies.

Long-term quality of life in inoperable non-small cell lung cancer patients treated with conventionally fractionated compared to hyperfractionated accelerated radiotherapy – Results of the randomized CHARTWEL trial

Background and purposeTo evaluate the quality of life (QoL) of patients with inoperable non-small cell lung cancer treated with conventionally fractionated radiotherapy (CF) vs. continuous hyperfractionated accelerated radiotherapy weekend-less (CHARTWEL).Material and methodsThe largest monocentric subgroup of the phase III CHARTWEL trial was analyzed up to three years after randomization. QoL was assessed with the European Organization for Research and Treatment of Cancer QoL Core Questionnaire (QLQ-C30) and lung cancer module (QLQ-LC13) and compared using linear mixed models. QoL interrelations with recurrence, metastasis, and death were explored by multi-state modeling.Results160 patients (98%) provided at least one QoL assessment. Average treatment differences of CF vs. CHARTWEL over three years were −5.4 points (95%CI [−13.6,2.8], p = 0.19) in global QoL, 11.9 ([2.8,21.0], p = 0.01) in fatigue, 13.4 ([3.5,23.3], p = 0.009) in pain, 10.5 ([1.3,19.6], p = 0.03) in dyspnea, and 5.2 ([−2.7,13.0], p = 0.19) in dysphagia. At 12 months, the probabilities of being disease-free with good, good or moderate, any global QoL, or alive were 5.1%, 20.3%, 34.2%, 54.4% under CF and 10.4%, 21.0%, 37.5%, 65.3% under CHARTWEL.ConclusionsOver three years, QoL was similar or more favorable under CHARTWEL compared to CF. Modeling QoL together with disease states provided additional insight into treatment comparisons.

Short communication: Effect of replacing corn with beet pulp in a high concentrate diet fed to weaned Holstein calves on diet digestibility and growth

Using soluble fiber sources in starter and grower feeds for dairy calves less than 4 mo of age is common to reduce costs compared with including traditional cereal grains. Beet pulp (BP) contains relatively high concentrations of pectin compared with other fibrous feed ingredients and has been shown to be an acceptable replacement for corn in adult cow diets. However, limited information is available on BP digestibility and growth performance for young calves fed diets with BP. In this study, 48 male Holstein calves (59 ± 2 d of age, 77 ± 2.2 kg of initial body weight) were fed 95% concentrate, 5% chopped grass hay diets in groups with 4 calves/pen for 56 d. Pens were randomly assigned to 1 of 3 dietary treatments containing 0, 15, or 30% BP on an as-fed basis. Body weights, hip widths, and body condition scores were assessed at 56 (start of trial), 84, and 112 d of age. Dry matter intakes and refusals were recorded daily by pen. Digestion coefficients (dC) of the diets and microbial protein flows were estimated when calves were approximately 84 d of age. Fecal samples were collected daily from pen floors over a 7-d period, and urine samples were collected from 2 calves/pen over a 2-d period and analyzed for purine derivatives. Calf average daily gain and hip width change decreased linearly (from 1.09 to 1.04 kg/d and 5.4 to 4.8 cm over 56 d, respectively) with increasing BP. Dry matter, organic matter (from 79.7 to 75.6%), crude protein (75.7 to 70.1%), and starch (97.1 to 93.1%) dC decreased with increasing inclusion rates of BP. Conversely, neutral detergent fiber (from 47.1 to 52.7%) and acid detergent fiber (44.1 to 53.0%) dC increased with increasing BP. Estimates of urine output and microbial protein flow using purine derivatives did not differ among treatments. Under the conditions of this study, BP reduced growth largely by reducing diet digestibility in dairy calves from 56 to 112 d of age.

Dry matter intake, body condition score, and grazing behavior of nonlactating, pregnant dairy cows fed kale or grass once versus twice daily during winter

The objective of this study was to examine the effect of wintering pregnant, nonlactating dairy cows outdoors on either kale or grass, fed in 1 [11 kg dry matter (DM) of kale or grass + 3 kg DM of baled barley straw offered in the morning] or 2 allocations (5.5 kg DM of kale or grass grazed + 1.5 kg DM of barley straw offered morning and afternoon) per day. The body condition score (BCS) gain over the 47-d winter feeding period was higher for grass-fed (0.5 BCS units) than kale-fed cows (0.3 BCS units), but was unaffected by feeding frequency. Forage DM utilization was higher for kale-fed (97%) than grass-fed cows (76%), leading to higher estimated dry matter intake (DMI) in kale-fed (10.7 kg of DM/cow per day) than grass-fed cows (7.7 kg of DM/cow per day). Forage DM utilization and estimated DMI were not affected by feeding frequency. Prehension bite rate was greater for grass-fed (37.3 bites/min) than kale-fed cows (7.6 bites/min), but more mastication bites were required for kale-fed cows. Cumulative DMI after 2, 3, and 6 h was greater in cows allocated forage once than twice a day and for kale than grass after 3 and 6 h. Mean eating time was greater for cows offered forage once (477 min) than twice (414 min) per day. In conclusion, increasing feeding frequency from once to twice per day decreased the intake rate within the first 6 h after allocation, but did not affect total daily DMI, DM utilization or BCS gain. Thus, moving cows more frequently would not have any significant advantage. It may increase labor requirements, thereby creating a more challenging wintering management than feeding once per day.

Association of rumination time and health status with milk yield and composition in early-lactation dairy cows

The objective of this study was to determine the associations of rumination time (RT) and health status with milk yield and milk composition. This study used 339 dairy cows from 4 commercial dairy farms in Ontario, Canada (first lactation, n = 107; second lactation, n = 112; ≥third lactation, n = 120). Rumination time was monitored (24 h/d) using an automated system from 1 to 28 d in milk (DIM). Cows were milked 3×/d on each farm, and 2 farms recorded milk weights at each milking to determine daily milk yield (n = 170). Cows were also monitored for milk composition (fat and protein content) 1×/wk. Last, subclinical ketosis (SCK) was diagnosed 1×/wk; cows with at least one blood sample with β-hydroxybutyrate ≥1.2 mmol/L postcalving were diagnosed with SCK. Cases of retained placenta, metritis, milk fever, or mastitis during the study period were also recorded. Cows were categorized into 1 of 4 groups: healthy cows that had no SCK or any other health issue (HLT; n = 139); cows that were treated for at least one health issue other than SCK (HLT+; n = 50); SCK cows with no other health problems during transition (HYK; n = 97); or cows that had SCK and one or more other health problems (HYK+; n = 53). All data were summarized by week across cows, and the associations between rumination time and milk yield (n = 170) and milk composition (n = 339) were modeled. Across all lactations, and including all health categories, milk yield increased by week, whereas fat and protein content both decreased by week. A positive association was found between summarized RT and milk yield in first-lactation (+0.006 ± 0.003 kg/min of RT) and second-lactation (+0.015 ± 0.004 kg/min of RT) cows from 4 to 28 DIM, as well as in ≥third-lactation cows; however, the relationship between RT and milk yield differed across weeks in those cows. A negative association between RT and milk fat content was found in ≥third-lactation cows (−0.002 ± 0.00059 percentage points/min of RT). From 4 to 28 DIM, ≥third-lactation HYK and HYK+ cows produced less protein (0.11 ± 0.051 and 0.13 ± 0.056 percentage points, respectively) than HLT cows. Over the 4-wk observation period, first-lactation HYK+ cows tended to deposit 0.11 ± 0.056 percentage points less protein in their milk compared with HLT cows. Second-lactation HYK+ cows produced less milk than HLT cows each week during early lactation. In summary, RT was positively associated with milk yield in early-lactation dairy cows, across all lactations, and negatively associated with milk fat content in ≥third-lactation cows. Further, the results showed that early-lactation cows that experience SCK, particularly with one or more other health problems, might have decreased milk yield and milk protein content.

Short communication: Genomic and phenotypic analyses of exopolysaccharides produced by Streptococcus thermophilus KLDS SM

Streptococcus thermophilus plays important roles in the dairy industry. Streptococcus thermophilus KLDS SM could produce a high amount of exopolysaccharides (EPS). To understand the possible link between the genotype and the phenotype regarding EPS, the complete genome of S. thermophilus KLDS SM was sequenced and investigated in silico for genes related to carbohydrate fermentation, nucleotide sugars synthesis, and EPS gene cluster. We found that S. thermophilus KLDS SM is able to ferment sucrose, mannose, glucose, galactose, and lactose from the genomic research, which was confirmed by API 50 CH (bioMérieux, Marcy l'Etoile, France). The genetic analysis of nucleotide sugars and EPS cluster revealed that the EPS produced by this strain are composed of galactose and glucose, in accordance with the biochemical result. Furthermore, differences in the molecular mass of EPS from S. thermophilus KLDS SM cultivated under different carbon sources were correlated with the transcription levels of the genes encoding chain length determination protein and glycosyltransferase. Our findings provide a better understanding of the link between the genetic elements and the chemical conformation of EPS and a theoretical basis for producing tailor-made EPS through genetic and metabolic engineering approaches.

Effects of Leymus chinensis replacement with whole-crop wheat hay on blood parameters, fatty acid composition, and microbiomes of Holstein bulls

This study investigated the replacement of Leymus chinensis (LC) with whole-crop wheat hay (WCWH) in the diets of Holstein bulls in the fattening stage and examined the potential effects on blood parameters, fatty acids in rumen fluid and serum, and the rumen microbiomes. In this study, 12 Holstein bulls in the fattening period (body weight = 485.0 ± 40.8 kg) were assigned to 1 of 4 dietary treatments using a 4 × 4 Latin square design. Each experimental period consisted of a 17-d adaptation period and a 5-d collection period. The dietary treatments consisted of 4 proportions of WCWH (0, 33, 67, and 100%) as a substitute for LC (designated as WCWH0, WCWH33, WCWH67, and WCWH100, respectively). On d 18 and 22 of each experiment period, blood and rumen content samples were collected for analysis, respectively. Real-time quantitative PCR was used to analyze the rumen microbiomes. The results from this study revealed no differences in the saturated, monounsaturated, and polyunsaturated long-chain fatty acid proportions of rumen liquid among the treatments. It was observed in the present trial that rumen microbiotal flora were not significantly different in the bulls fed LC compared with the bulls fed WCWH. Additionally, blood sample analysis demonstrated that the concentration of urea nitrogen in the WCWH100 group was higher than that observed in the other groups. Meanwhile, no differences were detected for other serum parameters. There were no differences in the proportions of serum saturated, monounsaturated, and polyunsaturated long-chain fatty acids among the treatments. In conclusion, our data revealed that LC can be replaced with WCWH in the diet of Holstein bulls in the fattening stage with no negative effects on the blood indicators, fatty acids, and microbiomes.

Short communication: Analysis of mycotoxins in Spanish milk

We surveyed the presence of 22 mycotoxins in 191 Spanish cow milk samples. Mycotoxins could be carried over from diet into animal milk and have toxic effects on human and animal health. The interaction of different mycotoxins may be additive or synergetic. Therefore, surveillance of mycotoxin co-occurrence in milk is recommended. Aflatoxins M1, B1, B2, G1, and G2, ochratoxins A and B, nivalenol, deoxynivalenol, deepoxy-deoxynivalenol, 3- and 15-acetyldeoxynivalenol, diacetoxyscirpenol, neosolaniol, fusarenon X, T-2 and HT-2 toxins, fumonisins B1, B2, and B3, sterigmatocystin, and zearalenone were analyzed. Samples were treated by liquid-liquid extraction with acidified acetonitrile, followed by an acetonitrile-water phase separation using sodium acetate. The analysis was carried out by HPLC coupled to a triple quadrupole mass spectrometer. None of the analyzed mycotoxins had a concentration level higher than their detection limit (0.05–10.1 µg/L). The aflatoxin M1 in the samples never exceeded the level established by the European Union.

Reusing reverse osmosis (RO) membrane permeate instead of potable water in the dairy industry is a very appealing tactic. However, to ensure safe use, the quality of reclaimed water must be guaranteed. To do this, qualitative and quantitative information about which compounds permeate the membranes must be established. In the present study, we provide a detailed characterization of ultrafiltration, RO, and RO polisher (ROP) permeate with regard to organic and inorganic compounds. Results indicate that smaller molecules and elements (such as phosphate, but mainly urea and boron) pass the membrane, and a small set of larger molecules (long-chain fatty acids, glycerol-phosphate, and glutamic acid) are found as well, though in minute concentrations (<0.2 µM). Growth experiments with 2 urease-positive microorganisms, isolated from RO permeate, showed that the nutrient content in the ROP permeate supports limited growth of 1 of the 2 isolates, indicating that the ROP permeate may not be guaranteed to be stable during protracted storage.

Growth performance, rumen fermentation, nutrient utilization, and metabolic profile of dairy heifers limit-fed distillers dried grains with ad libitum forage

The objective of this study was to determine the effects of feeding a corn- and soybean-product-based concentrate mix or distillers dried grains with solubles concentrate mix with ad libitum grass hay to dairy heifers. A 16-wk randomized complete block design study was conducted using 24 heifers [18 Holstein and 6 Brown Swiss; 219 ± 2 d of age (±standard deviation); 230 ± 4 kg of body weight] to evaluate the effect of diet on dry matter intake (DMI), growth performance, rumen fermentation, metabolic profile, and nutrient digestibility. Treatments were (1) corn and soybean product concentrate mix, and (2) distillers-dried-grains-with-solubles-based concentrate mix (DDG). Both concentrate mixes were limit-fed at 0.8% of body weight and grass hay was offered ad libitum. Heifers were individually fed using Calan gates and orts were recorded daily at feeding. Heifers were weighed every 2 wk and ration concentrate mix offered was adjusted accordingly. Frame measurements and body condition score were recorded every 2 wk. Rumen fluid was collected via esophageal tubing during wk 12 and 16 for pH, ammonia N, and volatile fatty acid analysis. Jugular blood samples were collected every 4 wk for metabolite and metabolic hormone analysis. Total-tract digestibility of nutrients was evaluated during wk 16 by fecal grab sampling. No treatment by week interactions were observed for any of the growth measurements and growth measurements and DMI did not differ between treatments. A treatment by time interaction was observed for rumen butyrate percentage with heifers fed DDG having a greater percentage. Total volatile fatty acid concentration, acetate molar percentage, and acetate:propionate decreased with the DDG treatment, whereas propionate molar percentage increased. No treatment by week interactions were observed for any of the metabolites or metabolic hormones measured. A tendency was observed for glucose and plasma urea nitrogen concentration to decrease with DDG. Plasma cholesterol and insulin increased with DDG. Results demonstrated that limit-feeding heifers DDG at 0.8% of body weight with ad libitum grass hay maintained growth performance, average daily gain, DMI, and gain:feed, with shifts in the metabolic profile compared with the corn and soybean product concentrate mix.

Transmission dynamics of intramammary infections caused by Corynebacterium species

The development of reliable models for transmission of intramammary infections (IMI) is the subject of extensive research. Such models are useful to enhance the identification and understanding of factors that affect pathogen-specific IMI dynamics. Longitudinal transmission models are valuable for predicting infection outbreak risks, quantifying the effectiveness of response tactics, and performing response planning. In this work, we focused on modeling Corynebacterium spp. by using a compartmental model. Previous investigations have considered modeling the transmission dynamics of several bacterial pathogens, but not Corynebacterium spp. We established a Corynebacterium spp. Susceptible–Infectious–Susceptible (SIS) model. We simulated the model numerically by using parameters that we estimated by a generalized linear model approach, using month of study as the time variable. The data, from which the parameters of the model were estimated, were obtained in a field trial conducted in 2 US dairy herds. Altogether, 786 cows were sampled at least once during the 13-mo study period. The total number of quarter milk cultures and cases of IMI caused by Corynebacterium spp. were 11,744 and 556, respectively, in farm A; the corresponding figures for farm B were 11,804 and 179. Our modeling study included only transmission from persistent IMI caused by Corynebacterium spp. within the lactation pens. The rate of new infections was significantly related to preexisting IMI in both farms, underscoring the importance of preexisting Corynebacterium spp. IMI for the transmission of Corynebacterium spp. within lactation pens. The estimated basic reproduction numbers (R0) in the 2 farms were 1.18 and 0.98, respectively. The nonsignificant disparity in R0 was associated with significant differences in cure rates between farms.

Describing temporal variation in reticuloruminal pH using continuous monitoring data

Reticuloruminal pH has been linked to subclinical disease in dairy cattle, leading to considerable interest in identifying pH observations below a given threshold. The relatively recent availability of continuously monitored data from pH boluses gives new opportunities for characterizing the normal patterns of pH over time and distinguishing these from abnormal patterns using more sensitive and specific methods than simple thresholds. We fitted a series of statistical models to continuously monitored data from 93 animals on 13 farms to characterize normal variation within and between animals. We used a subset of the data to relate deviations from the normal pattern to the productivity of 24 dairy cows from a single herd. Our findings show substantial variation in pH characteristics between animals, although animals within the same farm tended to show more consistent patterns. There was strong evidence for a predictable diurnal variation in all animals, and up to 70% of the observed variation in pH could be explained using a simple statistical model. For the 24 animals with available production information, there was also a strong association between productivity (as measured by both milk yield and dry matter intake) and deviations from the expected diurnal pattern of pH 2 d before the productivity observation. In contrast, there was no association between productivity and the occurrence of observations below a threshold pH. We conclude that statistical models can be used to account for a substantial proportion of the observed variability in pH and that future work with continuously monitored pH data should focus on deviations from a predictable pattern rather than the frequency of observations below an arbitrary pH threshold.
