Unveiling the impact of nanoparticle size dispersity on the behavior of polymer nanocomposites

Polymer nanocomposites (PNCs), a class of polymer materials incorporating nano-sized particles (NPs), have tremendous potential in industrial formulations and technological applications, such as protective coatings and food packaging. In this work, we perform Molecular Dynamics simulations to unveil the impact of NP size dispersity on a variety of properties that characterize the response of PNCs at the nano and macro scales. In particular, at the nano scale, we investigate the space distribution of small and large polydisperse NPs and their ability to diffuse through a dense isotropic distribution of unentangled polymer chains. We find very interesting scaling laws relating the average size and polydispersity index of NPs with their diffusion coefficients, generally underestimated by existing theoretical models. These theories are here adapted to include the effect of NP size dispersity and their predictions, confirming the relevance of incorporating such contributions, are validated against our simulation results. We also analyze the diffusivity of the polymer chains as a function of the interparticle distance for a spectrum of NP diameters and confirmed the existence of a single master curve as recently observed experimentally (S. Gam et al., Soft Matter, 2012, 8, 6512). To assess the effect of NP size dispersity on the macroscopic response of our model PNC, we evaluate two key transport properties, shear viscosity and thermal conductivity, which are found to display an intriguing universal behavior when plotted against the polymer/NP specific interface area and the inverse of the NP's mass, respectively.

A new strategy for fabricating polypropylene alloy with good low-temperature toughness was reported. Glass transition temperature (Tg) of rubber phase in polypropylene/ethylene-propylene rubber/poly(styrene-b-ethylene/propylene) diblock copolymer (PP/EPR/SEP) blend was continuously reduced through strengthening the interfacial tensile force on rubber phase by using the concept of mismatched thermal expansion coefficient. As a result, the brittle-tough transition (BTT) of PP alloy shifted to lower temperature and subsequently excellent impact strength at low temperature was achieved. Through qualitative analysis of impact force, it was found that the theoretical temperature at which BTT occurred was close to that obtained by experiment, indicating BTT of rubber toughened plastic system is indeed controlled by rubber's Tg at impact instant. Furthermore, the PP alloy with excellent low-temperature toughness shows balanced toughness-rigidity, on the contrary, the blends presents poor rigidity when SEP or EPR is used to toughen PP alone.

Pressure-sensitive behaviors, mechanisms and model of field assisted quantum tunneling composites

Field assisted quantum tunneling composite (FAQTC) is a unique pressure-sensitive material with the advantages of large resistance change range under external force, easy preparation and excellent mechanical properties. In this paper, with attentions to the pressure-sensitivity of the FAQTCs, the effects of silicon rubber matrix, diameter and dosage of nickel particles as well as magnetic field treatment are systematically investigated. The reproducibility of the pressure-sensitivity of the FAQTCs under cyclic load is explored. Based on Cotton's equation and Burger's model, the descriptions of stress relaxation behavior and electrical resistance relaxation behavior of the composites under static compressive loading are given respectively. The results show that external magnetic field during curing process allows better adjustment of the pressure-sensitivity of the FAQTCs with fewer nickel particles. The increase of the dosage of nickel particles can improve the stability and reproducibility of the pressure-sensitivity of the composites. Electrical resistance relaxation behavior of the composites is partly controlled by the stress relaxation behavior. Moreover, based on the theory of percolation conduction, the mechanism of the pressure-sensitivity of the FAQTCs under uniaxial load is discussed and further qualitatively explained by adopting effective conducting path model. Finally, on the basis of this model combined with quantum tunneling effect, a mathematical model describing the pressure-sensitivity of the composites is established, which can well describe the pressure-sensitivity of the composites.

Polyaniline film-based wireless photo reactor for hydrogen generation through exciton mediated proton reduction

The unique light absorption property of semiconductors has been exploited to build photovoltaic (PV), photo electrochemical solar cells (PESC) and photocatalytic (PC) reactors that not only convert sunlight into electrical energy but also drive useful chemical reactions. Hydrogen, the clean fuel of 21st century has been targeted for several decades. However, the above systems possess complex architecture with two or more semiconductor-solution interfaces that are often plagued with material instability and show poor photon conversion efficiency. Here we demonstrate a simple pristine polyaniline (PANI) film based photo H2 generating system. The 'wireless' system with only one PANI/solution interface, utilizes internal redox chemistry in the conjugated polymer for 'photo exciton' formation and charge separation. The electrons migrate to the PANI/aq. H2SO4 interface and reduce H+ to produce H2. The polymer film is light weight, flexible, and solution-processable. The reported system is simple and scalable for high volume production of hydrogen.

Crosslinked membranes of poly(vinyl ether)s having oxyethylene side chains: The effects of the side chain length and the crosslinkable group on CO2 permeability

Living cationic copolymerization of vinyl ethers having oxyethylene side chains (MOEO3VE and MOEO4VE) with vinyl ethers having thermal-crosslinkable groups (VEEM and VEEA) provided the random-like copolymers [poly (MOEO3VE-VEEM), poly (MOEO4VE-VEEM), poly (MOEO3VE-VEEA), and poly (MOEO4VE-VEEA)]. The obtained copolymer compositions were almost the same as those of the feed ratios, and the copolymers with various composition ratios were synthesized ([MOEO3VE or MOEO4VE]/[VEEM or VEEA] = 2:1, 4:1, 6:1, 10:1, and 14:1). All the copolymers are sticky liquid before crosslinking, but the relatively tough porous Teflon film-based composite membranes were obtained by thermal crosslinking via mathacrylate groups or acrylate groups. All the membranes showed high CO2 permselectivity (PCO2/PN2 = 40–51) due to the high CO2 solubility selectivity (SCO2/SN2 = 44–61). The CO2 permeability increased as the composition ratios of VEEM and VEEA decreased because the gas diffusivity enhanced by the decrement of crosslinking points. Given the same composition ratio, the increased oxyethylene side chain length is beneficial in enhancing CO2 permeability. The effects of crosslinkable groups on gas permeability were not observed. Poly (MOEO4VE-VEEM) and poly (MOEO4VE-VEEA) showed the excellent performance for CO2 separation (PCO2 = 440 and 460 barrer, PCO2/PN2 = 51).

Sustainable poly(ε-decalactone)−poly(l-lactide) multiarm star copolymer architectures for thermoplastic elastomers with fixed molar mass and block ratio

To improve the thermal and mechanical properties of thermoplastic elastomers derived from plant-based monomers, a series of well-defined multiarm star block copolymers was investigated. Controlled bulk ring-opening polymerization of ε-decalactone (DL) with multiarm initiators yielded hydroxyl-terminated (PDL−OH)n, which were subsequently converted to (PDL−PLLA)n using l-lactide (LLA) through a one-pot, two-step process. The multiarm copolymers were designed for targeting on Mn,NMR and fPLLA of 180 kg mol−1 and 0.27, respectively. Structural analysis proved that PLLA hard domains were thinner, and thus, more compact microphase-separated structures with hexagonally packed cylinders were induced by increasing n. Thermal and tensile property measurement demonstrated that, although the copolymers had a lower crystallinity in PLLA due to the shorter chain length, the thermal degradation stability and mechanical strength were enhanced, which was caused by to an amplified pinning effect caused by the more closed packed PLLA domains, in addition to chemical crosslinking of the PDL strands.

Guanidinium functionalized polypeptide nanogels as the phosphate binder

Phosphate-binding particles are clinically used to reduce the phosphate level in patients with chronic kidney diseases. However, the binding efficiency of the current amino-functional polymers is low, resulting in high pill burden and low patient adherence. Here, we used polylysine nanogels with amino- and guanidinium-functional groups (PLL nanogel and GPLL nanogel), respectively, and experimentally proved that guanidinium is a much more efficient phosphate-binding group than the amino group at all the tested conditions. The phosphate binding amount of the GPLL nanogel is 60% higher than that of the PLL nanogel at pH 7, 55% higher at pH 2, and 30% higher in the presence of 150 mM NaCl. This guanidination strategy should also be applicable to non-polypeptide systems for anion sequestration. Though the difference of amino and guanidinium functional groups interacting with phosphate ions had been known in the literature, this is the first experimental study to directly compare their phosphate binding performance in clinically relevant conditions (e.g., crosslinked particle forms with all the other parameters controlled to be the same). Therefore, guanidinium functionalized polymers have the potential to become the new generation therapeutics.

Tungstate-loaded triazine-based magnetic poly(Bis-imidazolium ionic liquid): An effective bi-functional catalyst for tandem selective oxidation/Knoevenagel condensation in water

A novel bi-functional polymeric catalyst was synthesized by immobilization of tungstate anions onto the nitrogen rich poly(ionic liquid)/magnetic nanocomposite. The resulting catalyst has two types of catalytic sites: (i) immobilized WO4 anions with bis-imidazolium ionic liquid cation for selective oxidation of alcohols and (ii) basic amine groups for Knoevenagel condensation between produced aldehyde and malononitrile. Due to the polymeric nature of the catalyst, large amounts of tungstate and basic nitrogen groups were presented on the solid surface which led to a decrease in the applied catalyst mass for catalytic reaction. High catalytic activity and excellent selectivity of catalyst in water medium make this protocol a green way for production of fine chemicals.

Complications and Cost Analysis of Intraoperative Arterial Complications in Head and Neck Free Flap Reconstruction

Background Microvascular anastomotic patency is fundamental to head and neck free flap reconstructive success. The aims of this study were to identify factors associated with intraoperative arterial anastomotic issues and analyze the impact on subsequent complications and cost in head and neck reconstruction. Methods A retrospective review was performed on all head and neck free flap reconstructions from 2005 to 2013. Patients with intraoperative, arterial anastomotic difficulties were compared with patients without. Postoperative outcomes and costs were analyzed to determine factors associated with microvascular arterial complications. A regression analysis was performed to control for confounders. Results Total 438 head and neck free flaps were performed, with 24 (5.5%) having intraoperative arterial complications. Patient groups and flap survival between the two groups were similar. Free flaps with arterial issues had higher rates of unplanned reoperations (p < 0.001), emergent take-backs (p = 0.034), and major surgical (p = 0.002) and respiratory (p = 0.036) complications. The overall cost of reconstruction was nearly double in patients with arterial issues (p = 0.001). Regression analysis revealed that African American race (OR = 5.5, p < 0.009), use of vasopressors (OR = 6.0, p = 0.024), end-to-side venous anastomosis (OR = 4.0, p = 0.009), and use of internal fixation hardware (OR =3.5, p = 0.013) were significantly associated with arterial complications. Conclusion Intraoperative arterial complications may impact complications and overall cost of free flap head and neck reconstruction. Although some factors are nonmodifiable or unavoidable, microsurgeons should nonetheless be aware of the risk association. We recommend optimizing preoperative comorbidities and avoiding use of vasopressors in head and neck free flap cases to the extent possible.

Equipping African American Clergy to Recognize Depression

Specificity protein 1 (Sp1) is a widely expressed transcription factor that plays an important role in the promotion of oncogenes required for tumor survival, progression and metastasis. Sp1 is highly expressed in several cancers including colorectal cancer (CRC) and is related to poor prognosis. Therefore, targeting Sp1 is a rational for CRC therapy. In this review, we will recapitulate the current understanding of Sp1 signaling, its molecular mechanisms, and its potential involvement in CRC growth, progression and metastasis. We will also discuss the current therapeutic drugs for CRC and their mechanism of action via Sp1.

Associations between trace elements and clinical health parameters in the North Pacific loggerhead sea turtle ( Caretta caretta ) from Baja California Sur, Mexico

Abstract

This study investigated selected trace elements toxicity in sea turtles Caretta caretta population from Baja California Sur (BCS), Mexico, by analyzing associations among Zn, Se, Cu, As, Cd, Ni, Mn, Pb, and Hg with various biochemical parameters (packed cell volume, leukocytes, and selected blood parameters), and whether their concentrations could have an impact on the health status of sea turtles. Blood samples from 22 loggerhead (C. caretta) sea turtles from BCS, Mexico, were collected for trace elements on biochemistry parameter analyses. Significant associations among trace element levels and the biochemistry parameters were found: Cd vs ALP (R 2 = 0.874, p ˂ 0.001), As vs ALP (R 2 = 0.656, p ˂ 0.001), Mn vs ALP (R 2 = 0.834, p ˂ 0.001), and Ni vs LDH (R 2 = 0.587, p ˂ 0.001). This study is the first report of the biochemical parameters of the North Pacific loggerhead sea turtle (C. caretta) from Baja California Sur, Mexico, and it is the first to observe several associations with toxic and essential trace elements. Our study reinforces the usefulness of blood for the monitoring of the levels of contaminating elements and the results suggest that, based on the associations with health clinical parameters, high levels of Cd and As could be representing a risk to the North Pacific loggerhead population health.

Also blood and kidney samples were collected for further laboratory and histological analyses.ResultsR. viburnoides leaves extract improved renal function. It also improved the renal function impairments caused by gentamicin-induced nephrotoxicity, as revealed by glomerular filtration rate, urine output and proteinuria.ConclusionR. viburnoides exert renoprotective effect, which may support its popular use for renal diseases treatment.

Ethnopharmacological relevanceIpomoea pes-caprae is known as bayhops, beach morning glory or goat's foot, and in Brazil as salsa-de-praia. Its leaves are used in worldwide folk medicine for the relief of jellyfish-stings symptoms. The literature only reports the neutralizing effects of nonpolar plant derived over jellyfish venoms, without validating the popular use or exploring the mechanism of action.Aim of the studyThis study aimed to evaluate and validate the topical effects of a semisolid containing hydroethanolic extract obtained from the aerial parts of I. pes-caprae using different models of paw- and ear-oedema and spontaneous nociception in mice, attempting to better understand the mechanism involved in its effect.Materials and methodsLeaf and stem of I. pes-caprae were extracted by ethanol 50% (v/v) and the soft-extract was incorporated in Hostacerin® cream base at 0.5%, 1.0% and 2% (w/w). The anti-hypersensitivity effects were evaluated by injecting the Physalia physalis venom into the right mice's hindpaw pre-treated either with the semisolid containing the I. pes-caprae extract or with the isolated majority compound 3,5-Di-O-caffeoylquinic acid (ISA). The topical anti-inflammatory activity was investigated using both preclinical models: paw oedema induced by trypsin, bradykinin (BK), histamine and carrageenan, and ear oedema induced by capsaicin. Additionally, the model of spontaneous nociception induced by trypsin and capsaicin were used to verify the topical antinociceptive activity.ResultsThe animals pre-treated with the semisolid containing I. pes-caprae extract or with the intraplantar injection of the major component (ISA) had the mechanical hypersensitivity induced by P. physalis venom significantly reduced. Significant inhibition was also observed in paw-oedema induced by trypsin, histamine and BK, and in a less extent in carrageenan-induced paw oedema. Similar effect was observed in mice challenged to the capsaicin-induced ear-oedema. Besides the vascular effects, the extract interfered with leukocyte migration induced by histamine injection. Finally, the semisolid presented significant inhibition in trypsin- and capsaicin-induced spontaneous nociception.ConclusionsThe hydroethanolic extract of I. pes-caprae showed compliance with the topical popular use of the herbal product to relieve the symptoms evoked by the cnidarian venom-skin contact, such as neurogenic oedema and nociception. The extract components seem to interfere with the effects resulting from the TRPV1, B2R and PAR-2 activation, once it interfered with painful-behaviour and oedema induced by capsaicin, BK and trypsin, pointing the histaminergic system as the main target, once it is an important mediator in the signalling pathway of the aforementioned receptors.

Ethnopharmacological relevanceSalvia miltiorrhiza polysaccharide (SMP) is one of the most important components in the water extract of Salvia miltiorrhiza Bunge, which has been mainly applied for the prevention or treatment of ischemic encephalopathy and cardiac diseases including myocardial infarction and coronary heart diseases in clinical practice.Aim of the studyOur object is to investigate the immune regulation effects of SMP, specifically on the proliferation and cytotoxicity of T lymphocytes through MAPK and NF-κB pathway in peripheral blood of cancer patients.Materials and methodsSMP was prepared through refluxing with ethanol, refluxing with water, Sevage treatment and ethanol precipitation. The lymphocytes were obtained from the peripheral blood of cancer patients. The effect of SMP on T lymphocyte proliferation was investigated by cell counting and flow cytometry. The effect of SMP on the proliferation of cancer cell lines A549, hepG2 and HCT116 was examined by MTT assay. The cytotoxic activity of T lymphocytes treated with SMP was detected by Calcein-acetoxymethyl (Calcein-AM) release. The gene expression of IL-4, IL-6, IFN-γ and toll like receptors (TLRs) was detected by semi-quantitative PCR. The protein expression of mitogen activated protein kinase (MAPK) and nuclear factor kappa-B (NF-κB) signaling pathway were detected by western blotting. To further verify whether SMP functions through the indicated pathways,, T lymphocytes were treated with SMP and an extracellular regulated protein kinase (ERK) inhibitor (U0126), a c-Jun N-terminal kinase (JNK) inhibitor (SP600125) or an inhibitor of NF-κB inhibitor-α (IκBα) (BAY11-7082), respectively. After 24 h co-treatment, the expressions of p-JNK, p-ERK, IκBα, inhibitory kappa B kinase α (IKKα) and inhibitory kappa B kinase β (IKKβ) protein were detected by western blotting, meanwhile cell numbers of T lymphocytes after inhibition were calculated again by cell counter.ResultsSMP dose-dependently promoted the proliferation of T lymphocytes of the cancer patients and significantly improved the cytotoxicity of T lymphocytes against cancer cells. However, SMP showed no effect on the proliferation of the tumor cells from the same source. Furthermore, the gene expression of cytokines including IL-4, IL-6 and IFN-γ were also up-regulated. Moreover, SMP enhanced gene expression of TLR1, TLR2 and TLR4; elevated protein expression of p-JNK and p-ERK; increased protein expression of IKKα, and IKKβ and decreased IκBα levels. Meanwhile, knockdown of ERK、JNK or IκBα expression with specific inhibitor significantly depressed the proliferation of T lymphocytes treated with SMP, corroborating the specific regulation effect of SMP on T lymphocytes through MAPK and NF-κB signaling pathways.ConclusionSMP specifically promotes the proliferation and enhances cytotoxicity of T lymphocytes in peripheral blood of cancer patients through activation of TLRs mediated -MAPK and -NF-κB signaling pathways.

Effects of the ethanol extract of black mulberry (Morus nigra L.) fruit on experimental atherosclerosis in rats

Ethnopharmacological relevanceAtherosclerosis (AS) is the major pathogenic component of coronary artery and cardiovascular disease. Studies have increasingly focused on natural medicines that have lipid-lowering, anti-inflammatory, and endothelial–protection activities. Black mulberry fruits are traditionally used in Uyghur folk medicine for the prevention and treatment of cardiovascular diseases in southern Xinjiang region of China. However, its underlying mechanisms remain unknown. Thus, our objective was to explore the effects and underlying mechanisms of ethanol extract of black mulberry (EEBM) in experimental atherosclerotic rats.Materials and methodsThe black mulberry fruit was extracted with acid ethanol and chromatographed on an AB-8 macroporous resin to obtain EEBM. Atherosclerotic rats were divided into five groups: normal, model, model plus simvastatin (5mg/kg·d·body weight), and model plus low-dose and high-dose EEBM groups (105 and 210mg/kg·d·body weight, respectively). Serum lipid profiles were measured by an automatic biochemistry analyser. The activities of anti-oxidative enzymes were determined using the chemical colorimetric method. Pathological changes in liver and arteries were examined by hematoxylin and eosin (H&E), and the intima-media thickness was measured.ResultsThe high-dose EEBM group showed significantly reduced total cholesterol, triglyceride, and low-density lipoprotein-cholesterol levels, as well as atherogenic index. Furthermore, treatment with high-dose EEBM markedly decreased malondialdehyde content and enhanced anti-oxidative enzyme activities. Histopathological examination showed that EEBM attenuated hepatic steatosis and reduced intima-media thickness and arterial atherosclerotic lesions in atherosclerotic rats. These results suggest that EEBM suppressed atherosclerosis development in atherosclerotic rats by regulating lipid metabolism abnormalities, enhancing anti-oxidative activities, and reducing atherosclerotic lesions, which could be attributed to anthocyanins (23.75%), or the cooperative action of anthocyanins, polyphenols (2.95%), and flavonoids (0.94%).

Analgesic and anti-inflammatory activities of 80% methanol root extract of Jasminum abyssinicum Hochst. ex. Dc. (Oleaceae) in mice

BackgroundPain and inflammation are associated with the pathophysiology of various clinical conditions. Most analgesic and anti-inflammatory drugs available in the market present a wide range of problems. The current study was aimed at investigating the analgesic and anti-inflammatory activity of 80% methanol extract of J.abyssinicum root.MethodsThe analgesic activity was determined using tail-flick test and acetic acid induced writhing, whereas anti-inflammatory activity was determined by carrageenan induced paw edema and formalin induced pedal edema, carried out in vivo. The test group received three different doses of the extract (50mg/kg, 100mg/kg and 200mg/kg) orally. The positive control group received diclofenac (10mg/kg), aspirin (100mg/kg or 150mg/kg) or morphine (20mg/kg) orally. The negative control group received vehicle (2% Tween 80, 10ml/kg) orally. Furthermore, preliminary phytochemical screening was carried out.ResultsOral administration of J.abbysinicum 80% methanol extract (at all doses) significantly (p < 0.001) inhibit pain sensation in the pain models. Similarly, the extract demonstrated anti-inflammatory effect in the inflammation models in mice. Preliminary phytochemical screening showed the presence of saponins, flavonoids, terepenoids, triterpenens and glycosides.ConclusionThe data obtained from the present study indicates that the extract possessed a significant analgesic and anti-inflammatory activity, upholding the folkloric use of the plant.

Endothelial cells (ECs) provide angiocrine factors orchestrating tumor progression. Here, we show that activated Notch1 receptors (N1ICD) are frequently observed in ECs of human carcinomas and melanoma, and in ECs of the pre-metastatic niche in mice. EC N1ICD expression in melanoma correlated with shorter progression-free survival. Sustained N1ICD activity induced EC senescence, expression of chemokines and the adhesion molecule VCAM1. This promoted neutrophil infiltration, tumor cell (TC) adhesion to the endothelium, intravasation, lung colonization, and postsurgical metastasis. Thus, sustained vascular Notch signaling facilitates metastasis by generating a senescent, pro-inflammatory endothelium. Consequently, treatment with Notch1 or VCAM1-blocking antibodies prevented Notch-driven metastasis, and genetic ablation of EC Notch signaling inhibited peritoneal neutrophil infiltration in an ovarian carcinoma mouse model.

Psychological distress and parent reporting on child health: The case of developmental delay

BackgroundCaregiver-completed screening questionnaires are a common first step in the identification of developmental delay. A caregiver's mood and anxiety level, however, may affect how he or she perceives and reports possible problems.AimsIn this article, we consider the association between caregiver distress and the accuracy of the Ages and Stages Questionnaire (ASQ), a widely-used screen.Methods and proceduresOur sample includes 857 parent-child dyads drawn from the Psychometric Assessment of the NDDS Study (PANS) and the NDDS Alternate Responses Study (NARS). Parents completed the ASQ and the K6, a brief measure of generalized distress. Children were assessed using the Bayley Scales of Infant and Child Development (BSID). We divided children on BSID result and used logistic regression to examine how distress influenced the ASQ's accuracy in each group.ResultsOf our 857 children, 9% had at least one domain below −2 standard deviations on the BSID, and 17.3% had positive ASQ results. Caregiver distress predicted a positive ASQ substantially and significantly more strongly among BSID-positive children than among others. This translates into slightly reduced ASQ specificity but greatly improved sensitivity among caregivers with higher distress.ConclusionsAt low to moderate levels of distress, greater distress is associated with greater ASQ accuracy.

Risk of second primary cancer associated with pre-diagnostic smoking, alcohol, and obesity in women with keratinocyte carcinoma

Keratinocyte carcinoma (KC), which includes basal-cell carcinoma (BCC) and squamous-cell cancer (SCC), has been associated with an increased risk of second primary cancers (SPCs), although the reason for this increase is unknown. We assessed the effects of smoking, alcohol, and obesity prior to the diagnosis of KC on the development of SPCs, as these are well-established risk factors for multiple cancers and may also contribute to the increased risk of SPCs among those with KC. A total of 15,628 women with self-reported KC were identified in the Nurses' Health Study. Incident SPCs were assessed throughout the follow-up until June 2012. Cox proportional hazards models were used to calculate the hazard ratios (HRs) of SPC associated with pre-diagnostic smoking, alcohol and body mass index (BMI). We also compared these risk estimates to those for first cancers in all cohort participants. During 193,695 person-years of follow-up, we recorded 2839 SPC cases. Compared with never smokers, current smokers had a significantly elevated risk for SPC overall and specifically for lung, colorectal, and bladder cancers. We also found a positive association between higher BMI and risk for SPC overall as well as for endometrial and bladder SPCs. Women with KC who consumed alcohol ≥30g/day had a marginally higher risk of SPC compared to non-drinkers. The associations between incident SPC risk among KC cases and smoking, alcohol, and obesity appeared similar to the associations between these risk factors and the incident first primary cancers in the whole cohort. Only in the heavy smoking (≥25 cigarettes/day) category was the HR for SPC after KC (2.34; 95% CI 1.98–2.76) slightly higher than that for the first cancer in the overall cohort (HR 1.86; 95% CI 1.75–1.98, Pheterogeneity=0.01). In conclusion, pre-diagnostic smoking, alcohol and obesity prior to KC diagnosis were associated with risk of SPCs.

BackgroundThe injection of amphetamine is becoming increasingly common. However, there has been a lack of research examining people who inject amphetamine as the primary drug of use, limiting the potential to ensure services address the unique needs of this group. The current study used latent class analysis to identify classes of polydrug use among people who report injecting amphetamine during the past 12months. It also examined differences between classes and drug use patterns, injecting practices, quality of life and psychological distress.MethodsParticipants who were attending non-government specialist alcohol and other drug treatment across New South Wales, Australia and had identified amphetamine as their principle drug of concern and reported injecting amphetamine in the previous 12months were included in the current study (N=827). Latent class analysis was performed to identify polydrug profiles of participants.ResultsThe large majority of people in the current study (85%) demonstrated low probability of heroin or other opiate use. Three distinct classes of polydrug use were identified: (1) Low-polydrug (n=491), (2) Opiates-polydrug (n=123), and (3) Alcohol-polydrug (n=213). There was a trend for the Low-polydrug class to demonstrate better functioning and safer injecting practices than the Opiates-polydrug and Alcohol-polydrug classes.ConclusionThe results suggest that the majority of people accessing treatment who inject amphetamine as their primary drug of choice have a low probability of heroin or other opiate use. It is important that future research consider whether traditional harm minimisation strategies are appropriate for people who primarily inject amphetamine.

Perceived risk and benefits of e-cigarette use among college students

Recent data demonstrates that the use of e-cigarettes is growing, especially among college students and young adults. This trend is increasingly problematic, as many of these individuals report never using traditional tobacco cigarettes, but nevertheless are using e-cigarettes. The present study sought to develop the Risks and Benefits of E-cigarettes (RABE) questionnaire to assess the perceptions about e-cigarette use among college students. College students (N=734) completed the RABE via online survey. Principal components analysis yielded two reliable scales representing perceptions about e-cigarette use. Based on the two-factor solution, subscales were named according to item content. The resulting 30 items demonstrated excellent internal consistency (Risks scale α=0.92; Benefits scale α=0.89). Subsequent confirmatory factor analysis generally supported the 2-factor structure. As an initial measure of construct validity, scale scores were compared across smoking status groups. Smoking status groups were defined by the following: "e-cigarette users" were current daily users of e-cigarettes, "conventional smokers" were daily traditional cigarette users, and "dual users" were individuals who used both e-cigarettes and traditional cigarettes daily. Scale scores for perceived Benefits of e-cigarette use differed significantly across groups (p<0.001), whereby students who reported using e-cigarettes or traditional cigarettes reported benefits associated with e-cigarette use. Scale scores for perceived Risks of e-cigarette use across smoking status groups did not significantly differ. The present results indicate that the RABE is a reliable instrument to measure college student's perceived risks and benefits of e-cigarettes.

Streptococcus pneumoniae is a serious Gram-positive pathogen that can lead to an invasive pneumococcal disease with high mortality rate. Pneumococcal capsular polysaccharide (PS) is a key virulence determinant and its immunogenicity can be increased by conjugation with a carrier protein. However, the PS-specific cellular and humoral immunity of pneumococcal conjugate vaccine needs further improvement. Hexadecane (HD) is an element of lipid that decorates the surface of nearly all microbial classes. Polyethylene glycol (PEG)-HD conjugate (PEG-HD) is soluble and can act as an adjuvant. In the present study, a novel pneumococcal polysaccharide conjugate vaccine was prepared by conjugation of tetanus toxoid (TT) portion of PS-TT conjugate (PS-TT) with PEG-HD. As compared with PS-TT, conjugation with PEG-HD led to an 8.0-fold increase in the PS-specific IgG titers. Conjugation with PEG-HD also gave rise to 34.9-, 3.6- and 7.7-fold increase in the IFN-γ, TNF-α and IL-5 levels, respectively. Thus, the conjugated PEG-HD has a stimulatory adjuvant activity to potentiate a robust humoral and cellular immunity. Our proposed conjugate was expected to act as an effective pneumococcal conjugate vaccine for prevention of S. pneumoniae infections.

Interplay between SIRT1 and hepatitis B virus X protein in the activation of viral transcription

Hepatitis B virus (HBV) genome is organized into a minichromosome known as covalently closed circular DNA (cccDNA), which serves as the template for all viral transcripts. SIRT1 is an NAD+-dependent protein deacetylase which activates HBV transcription by promoting the activity of cellular transcription factors and coactivators. How SIRT1 and viral transactivator X protein (HBx) might affect each other remains to be clarified. In this study we show synergy and mutual dependence between SIRT1 and HBx in the activation of HBV transcription. All human sirtuins SIRT1 through SIRT7 activated HBV gene expression. The steady-state levels of SIRT1 protein were elevated in HBV-infected liver tissues and HBV-replicating hepatoma cells. SIRT1 interacted with HBx and potentiated HBx transcriptional activity on precore promoter and covalently closed circular DNA (cccDNA) likely through a deacetylase-independent mechanism, leading to more robust production of cccDNA, pregenomic RNA and surface antigen. SIRT1 and HBx proteins were more abundant when both were expressed. SIRT1 promoted the recruitment of HBx as well as cellular transcriptional factors and coactivators such as PGC-1α and FXRα to cccDNA. Depletion of SIRT1 suppressed HBx recruitment. On the other hand, SIRT1 recruitment to cccDNA was compromised when HBx was deficient. Whereas pharmaceutical agonists of SIRT1 such as resveratrol activated HBV transcription, small-molecule inhibitors of SIRT1 including sirtinol and Ex527 exhibited anti-HBV activity. Taken together, our findings revealed not only the interplay between SIRT1 and HBx in the activation of HBV transcription but also new strategies and compounds for developing antivirals against HBV.

Kinetics of nucleotide entry into RNA polymerase active site provides mechanism for efficiency and fidelity

During transcription, RNA polymerase II elongates RNA by adding nucleotide triphosphates (NTPs) complementary to a DNA template. Structural studies have suggested that NTPs enter and exit the active site via the narrow secondary pore but details have remained unclear. A kinetic model is presented that integrates molecular dynamics simulations with experimental data. Previous simulations of trigger loop dynamics and the dynamics of matched and mismatched NTPs in and near the active site were combined with new simulations describing NTP exit from the active site via the secondary pore. Markov state analysis was applied to identify major states and estimate kinetic rates for transitions between those states. The kinetic model predicts elongation and misincorporation rates in close agreement with experiment and provides mechanistic hypotheses for how NTP entry and exit via the secondary pore is feasible and a key feature for achieving high elongation and low misincorporation rates during RNA elongation.

Targeting telomerase and telomeres to enhance ionizing radiation effects in in vitro and in vivo cancer models

One of the hallmarks of cancer consists in the ability of tumor cells to divide indefinitely, and to maintain stable telomere lengths throughout the activation of specific telomere maintenance mechanisms (TMM). Therefore in the last fifteen years, researchers proposed to target telomerase or telomeric structure in order to block limitless replicative potential of cancer cells providing a fascinating strategy for a broad-spectrum cancer therapy.In the present review, we report in vitro and in vivo evidence regarding the use of chemical agents targeting both telomerase or telomere structure and showing promising antitumor effects when used in combination with ionizing radiation (IR). RNA interference, antisense oligonucleotides (e.g., GRN163L), non-nucleoside inhibitors (e.g., BIBR1532) and nucleoside analogs (e.g., AZT) represent some of the most potent strategies to inhibit telomerase activity used in combination with IR. Furthermore, radiosensitizing effects were demonstrated also for agents acting directly on the telomeric structure such as G4-ligands (e.g., RHPS4 and Telomestatin) or telomeric-oligos (T-oligos). To date, some of these compounds are under clinical evaluation (e.g., GRN163L and KML001).Advantages of Telomere/Telomerase Targeting Compounds (T/TTCs) coupled with radiotherapy may be relevant in the treatment of radioresistant tumors and in the development of new optimized treatment plans with reduced dose adsorbed by patients and consequent attenuation of short- end long-term side effects. Pros and cons of possible future applications in cancer therapy based on the combination of T/TCCs and radiation treatment are discussed.

Quantifying trace elements in the emitted particulate matter during cooking and health risk assessment

Abstract

Particulate matter (PM) measurements were conducted during heating corn oil, heating corn oil mixed with the table salt and heating low fat ground beef meat using a PTFE-coated aluminum pan on an electric stove with low ventilation. The main objectives of this study were to measure the size segregated mass concentrations, emission rates, and fluxes of 24 trace elements emitted during heating cooking oil or oil with salt and cooking meat. Health risk assessments were performed based on the resulting exposure to trace elements from such cooking activities. The most abundant elements (significantly different from zero) were Ba (24.4 ug m−3) during grilling meat and Ti during heating oil with salt (24.4 ug m−3). The health assessment indicates that the cooking with an electric stove with poor ventilation leading to chronic exposures may pose the risk of significant adverse health effects. Carcinogenic risk exceeded the acceptable level (target cancer risk 1 × 10−6, US EPA 2015) by four orders of magnitude, while non-carcinogenic risk exceeded the safe level (target HQ = 1, US EPA 2015) by a factor of 5–20. Cr and Co were the primary contributors to the highest carcinogenic and non-carcinogenic risks, respectively.

Analysis of certain fatty acids and toxic metal bioaccumulation in various tissues of three fish species that are consumed by Turkish people

Abstract

Concentrations of toxic metals (Mn, Ni, Hg, Cd, Pb, Cr) in the muscle, skin, and liver of Mugil cephalus, Mullus barbatus, and Pagellus erythrinus which were purchased in large supermarkets of Elazig, and Mullus barbatus, which were caught on the sea of İskenderun Bay, Turkey, were analyzed. Fundamental analyses were carried out by inductively coupled plasma-mass spectrometry (ICP-MS) after samples were prepared by microwave digestion. Mean metal concentrations in different tissues were varied in the ranges of Cd 4–426, Cr 116–4458, Mn 141–24774, Hg 9–471, Pb 96–695, and Ni 68–6581 μg kg−1, for wet weight. The investigated metal bioaccumulation in the muscles of fish species, in general, was lower than those in the liver and skin. This method was verified by NCS ZC73016 chicken trace element-certified reference material analysis. In addition, fatty acids in the muscles of three fish species were analyzed. According to the gas chromatography (GC) results of fatty acids, the monounsaturated fatty acids (MUFA) were found to be between 23.76 and 31.97%. The fatty acids' polyunsaturated fatty acids (PUFA) ratio was found to be between 13.67 and 30.71% and saturated fatty acids ratios were determined in the range of 24.06–32.30%. In all fish species, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) ratio, which increase the value of these fish species, were high. These results show that these three fish species are good sources of fatty acids.

Removal of heavy metals from polluted soil using the citric acid fermentation broth: a promising washing agent

Abstract

The citric acid fermentation broth was prepared and it was employed to washing remediation of heavy metal-polluted soil. A well-defined washing effect was obtained, the removal percentages using citric acid fermentation broth are that 48.2% for Pb, 30.6% for Cu, 43.7% for Cr, and 58.4% for Cd and higher than that using citric acid solution. The kinetics of heavy metals desorption can be described by the double constant equation and Elovich equation and is a heterogeneous diffusion process. The speciation analysis shows that the citric acid fermentation broth can effectively reduce bioavailability and environmental risk of heavy metals. Spectroscopy characteristics analysis suggests that the washing method has only a small effect on the mineral composition and does not destroy the framework of soil system. Therefore, the citric acid fermentation broth is a promising washing agent and possesses a potential practical application value in the field of remediation of soils with a good washing performance.

Indoor air quality in public utility environments—a review

Abstract

Indoor air quality has been the object of interest for scientists and specialists from the fields of science such as chemistry, medicine and ventilation system design. This results from a considerable number of potential factors, which may influence the quality of the broadly understood indoor air in a negative way. Poor quality of indoor air in various types of public utility buildings may significantly affect an increase in the incidence of various types of civilisation diseases. This paper presents information about a broad spectrum of chemical compounds that were identified and determined in the indoor environment of various types of public utility rooms such as churches, museums, libraries, temples and hospitals. An analysis of literature data allowed for identification of the most important transport paths of chemical compounds that significantly influence the quality of the indoor environment and thus the comfort of living and the health of persons staying in it.

Oxygen microprofiles within the sediment-water interface studied by optode and its implication for aeration of polluted urban rivers

Abstract

To reveal the detailed vertical oxygen distribution at the sediment-water interface (SWI) and its relation with the oxygen consumption processes during and after aeration of polluted urban rivers, experimental systems constructed with collected sediment and in situ overlying water from a polluted urban river were aerated above or beneath the sediment-water interface 12 h a day for 15 days and left nonaerated for the following 10 days. The results showed that aeration of water or sediment both increased dissolved oxygen (DO) concentrations in the SWI, characterized by shifts in a "decrease-increase-decrease" manner during around 3 h for the aeration of water treatment (AW) and 6 h for the aeration of sediment treatment (AS). The oxygen penetration depth for AS experiments was between 0.66 and 4.16 mm with an average of 1.79 mm, significantly higher than that for AW experiments; however, the oxygen dissipation constant (mm−1) measuring the decay rate of DO near the SWI was greater for the AW experiments than the AS experiments. During the 10-day nonaeration period, the accumulation of nitrate in both the overlying water and sediment was greatly increased concomitantly with the higher oxygenation in AS experiments. From the nitrogen removal viewpoint, these results suggest that the SWI needs moderate oxygenation which enables nitrate and nitrite to be removed by denitrification rather than to be totally nitrified and accumulate as would result from the conventional practice by singly elevating DO concentrations.

Evaluation of black crust formation and soiling process on historical buildings from the Bilbao metropolitan area (north of Spain) using SEM-EDS and Raman microscopy

Abstract

In the present work, several building materials suffering from black crusts and soiled surfaces were evaluated by scanning electron microscopy energy dispersive X-ray spectrometry (SEM-EDS) and micro-Raman spectroscopy. The goal was to examine the elemental and molecular composition, the distribution on the samples, and the morphology of endogenous and exogenous compounds on those black crusts and soiled surfaces. The black crusts were deposited over different building materials such as limestone, sandstone, and brick that constitute a small construction called "malacate" as well as over a limestone substrate of a cemetery gate. Both constructions are dated back to the beginning of the twentieth century. The samples of soiling were taken from the façade of a building constructed in the 1980s. The analytical evaluation allowed in a first stage the determination of the composition and the observation of the morphology of soiling and black crusts. In addition, the evaluation of the compositions of the soiling and black crusts of different grade and formation allowed the assessment of the main weathering phenomena that the buildings have suffered, which were found to be sulfate impact, marine aerosol impact, depositions of metallic particles, crustal particulate matter depositions, carbonaceous particles, biodeterioration, and vandalism.

The Effect of Teaching Search Strategies on Perceptual Performance

Rationale and ObjectivesRadiology expertise is dependent on the use of efficient search strategies. The aim of this study is to investigate the effect of teaching search strategies on trainee's accuracy in detecting lung nodules at computed tomography.Materials and MethodsTwo search strategies, "scanning" and "drilling," were tested with a randomized crossover design. Nineteen junior radiology residents were randomized into two groups. Both groups first completed a baseline lung nodule detection test allowing a free search strategy, followed by a test after scanning instruction and drilling instruction or vice versa. True positive (TP) and false positive (FP) scores and scroll behavior were registered. A mixed-design analysis of variance was applied to compare the three search conditions.ResultsSearch strategy instruction had a significant effect on scroll behavior, F(1.3) = 54.2, P < 0.001; TP score, F(2) = 16.1, P < 0.001; and FP score, F(1.3) = 15.3, P < 0.001. Scanning instruction resulted in significantly lower TP scores than drilling instruction (M = 10.7, SD = 5.0 versus M = 16.3, SD = 5.3), t(18) = 4.78, P < 0.001; or free search (M = 15.3, SD = 4.6), t(18) = 4.44, P < 0.001. TP scores for drilling did not significantly differ from free search. FP scores for drilling (M = 7.3, SD = 5.6) were significantly lower than for free search (M = 12.5, SD = 7.8), t(18) = 4.86, P < 0.001.ConclusionsTeaching a drilling strategy is preferable to teaching a scanning strategy for finding lung nodules.

Cardiovascular Computed Tomography Findings after Pneumonectomy

Rationale and ObjectivesTo identify and compare cardiovascular findings on computed tomography (CT) scans after pneumonectomy (PNX) with those after lobectomy (LOBX).Materials and MethodsPre- and postoperative CT scans from 25 PNX patients were retrospectively analyzed and compared to those from 30 LOBX patients. The diameter of the main pulmonary artery (PA) and its ratio to the ascending aorta (PA/Ao) were determined. Cardiac morphometry values were ascertained by measuring maximum diameters of the right and left ventricle on axial (RVaxial, LVaxial) and four-chamber (RV4-ch, LV4-ch) views. RVaxial/LVaxial and RV4-ch/LV4-ch ratios were calculated. Vessel stumps were evaluated for thrombosis.ResultsAfter PNX, PA (31.1 ± 5.8 mm vs 28.7 ± 5.4 mm, P = 0.003), PA/Ao (0.97 ± 0.15 vs 0.86 ± 0.12, P = 0.0001), and cardiac morphometry values significantly increased (RVaxial 43.6 ± 7.4 vs 39.4 ± 7.1, P = 0.029; RV4-ch 41.1 ± 6.3 vs 37.6 ± 5.7, P = 0.041; RVaxial/LVaxial 1.18 ± 0.27 vs 1.03 ± 0.22, P = 0.04; RV4-ch/LV4-ch 1.17 ± 0.21 vs 1.02 ± 0.16, P = 0.03). There were no significant differences between right and left PNX. One case of PA stump thrombosis was identified after right PNX. LOBX resulted in a significant increase in PA (30.6 ± 4.3 vs 28.7 ± 3.5, P = 0.005) and PA/Ao (0.90 ± 0.09 vs 0.85 ± 0.10, P = 0.017), whereas cardiac morphometry values were not significantly changed compared to baseline values. No vessel stump thrombosis was observed after LOBX. In comparison to LOBX, all ascertained values were significantly elevated after PNX.ConclusionsMorphologic alterations of the cardiovascular system following PNX can be identified on CT scans. Alterations are more distinct after PNX compared to LOBX.

An Analysis of Future Publications, Career Choices, and Practice Characteristics of Research Presenters at an American College of Surgeons State Conference: A 15-Year Review

ObjectivesThe Accreditation Council for Graduate Medical Education requires scholarly activity within general surgery residency programs. The association between in-training research presentations and postgraduation publications is unknown. We hypothesized that surgical trainee presentations at an American College of Surgeons (ACS) state chapter meeting resulted in peer-reviewed publications and future scholarly activity.DesignThe ACS Wisconsin state chapter meeting agendas from 2000 to 2014 were reviewed to identify all trainees who delivered podium presentations. A literature search was completed for subsequent publications. Program coordinators were queried and an electronic search was performed to determine practice location and type for each residency graduate.SettingWisconsin state chapter ACS meeting.ParticipantsGeneral surgery residents, fellows, and medical students in Wisconsin.ResultsThere were 288 podium presentations by trainees (76% residents, 20% medical students, and 4% fellows). Presentations were clinical (79.5%) and basic science (20.5%). There were 204 unique presenters; 25% presented at subsequent meetings. Of these unique presenters, 46% published their research and 31% published additional research after residency. Among presenters who completed residency or fellowship (N = 119), 34% practiced in a university setting, and 61% practiced in a community setting; 31% practiced in Wisconsin. When comparing clinical vs basic science presenters, there was no difference in fellowship completion (37% vs 44%; p = 0.190) or practice type (38% vs 46% in a university setting; p = 0.397). Repeat presenters were more likely to pursue a fellowship vs those presenting once (76% vs 37%; p = 0.001).ConclusionsResearch presentations by surgical trainees at an ACS state chapter meeting frequently led to peer-reviewed publications. Presenters were likely to pursue research opportunities after residency. Repeat presenters were more likely to pursue a fellowship. ACS Wisconsin chapter meetings provide an excellent opportunity for scholarly activity. These outcomes should encourage ACS chapters and ACS members to support trainee research.

Vitamin D status is usually assessed by measuring the serum 25-hydroxyvitamin D (25(OH)D) concentration. There has been a dramatic increase in 25-OHD requests over recent years prompting many laboratories to consider the use of automated immunoassays. In this presentation, we will discuss and compare the two major techniques that are used for measuring of vitamin D (the binding assay and chemical assay techniques).Chemiluminescence immunoassays (CLIA), radioimmunoassy (RIA), and binding protein assay are belonging to the binding assay, while the chemical assay includes high performance liquid chromatography (HPLC) and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Significant differences in the 25(OH)D determination were observed between various assays. Standardization and harmonization of 25(OH)D measurements are therefore urgently needed. The widespread introduction of well standardized assays in clinical laboratories is the challenge in the next years.

Composition and mixing states of brown haze particle over the Himalayas along two transboundary south-north transects

Pollutants that are usually transported from southern Asia to the Tibetan Plateau deposit on the Plateau surface, change snow albedo and thereby surface radiative flux. This results numerous climatic implications like as erratic monsoon, perturbation in hydrological cycle, etc. However, the accurate estimation of these climatic implications is not well understood, because the atmospheric pollution is a heterogeneous mixture of various particle types. Therefore, this part of climate research requires a detailed investigation of physical and chemical properties of atmospheric pollutants. This study aimed to examine the physical and chemical properties of atmospheric pollutants across the Himalayan regions along two transboundary south-north transects. The information of individual-particles was obtained using microscopy-based techniques that comprises transmission electron microscope (TEM) and Energy-dispersive X-ray spectrometer (EDX). Study capture the signatures of various types of atmospheric species such as black carbon (BC), mineral dust, fly ash, organic matter, sulfate, nitrite, ammonium, and NaCl. Microscopy-based techniques confirm that these particles were generally in mixing state, for example salt-coated particles accounting for 25–56% of the total particles in sampled locations. Our analysis shows that urban and rural locations are characterized with atmospheric particles which sourced from anthropogenic activities, whereas remote locations with those released from natural crustal. However, the relative contributions of anthropogenic particles were higher than that of particles released from natural crustal. The presence of such particles over remote locations of Himalayan region provides an evidence of prevailing atmospheric transport processes, which further need to be well understood. It is expected that this work would be helpful in understanding the regional atmospheric conditions and the transboundary transport process of haze particles. As these informations are of great importance in modeling studies, which further lead to improve understanding of haze particles climate effects.

Soil and vegetation-atmosphere exchange of NO, NH3, and N2O from field measurements in a semi arid grazed ecosystem in Senegal

The alternating between dry and wet seasons and the consecutive microbial responses to soil water content in semiarid ecosystems has significant consequences on nitrogen exchanges with the atmosphere. Three field campaigns were carried out in a semi arid sahelian rangeland in Dahra (Ferlo, Senegal), two at the beginning of the wet season in July 2012 and July 2013, and the third one in November 2013 at the end of the wet season. The ammonia emission potentials of the soil ranged from 271 to 6628, indicating the soil capacity to emit NH3. The ammonia compensation point in the soil ranged between 7 and 150 ppb, with soil temperatures between 32 and 37 °C. Ammonia exchange fluctuated between emission and deposition (from −0.1–1.3 ng N.m−2 s−1), depending on meteorology, ambient NH3 concentration (5–11 ppb) and compensation point mixing ratios. N2O fluxes are supposed to be lower than NO fluxes in semi arid ecosystems, but in Dahra N2O fluxes (5.5 ± 1.3 ng N m−2 s−1 in July 2013, and 3.2 ± 1.7 ng N m−2 s−1 in November 2013) were similar to NO fluxes (5.7 ± 3.1 ng N m−2 s−1 in July 2012, 5.1 ± 2.1 ng N m−2 s−1 in July 2013, and 4.0 ± 2.2 ngN m−2 s−1 in November 2013). Possible reasons are the influence of soil moisture below the surface (where N2O is produced) after the beginning of the wet season, the potential aerobic denitrification in microsites, the nitrifier denitrification, and nitrification processes. The presence of litter and standing straw, and their decomposition dominated N compounds emissions in November 2013, whereas emissions in July 2012 and 2013, when the herbaceous strata was sparse, were dominated by microbial processes in the soil. CO2 respiration fluxes were high in the beginning (107 ± 26 mg m−2 h−1 in July 2013) and low in the end of the wet season (32 ± 5 mg m−2 h−1 in November 2013), when autotrophic and heterotrophic activity is reduced due to low soil moisture conditions These results confirm that contrasted ecosystem conditions due to drastic changes in water availability in semi arid regions have important non linear impacts on the biogeochemical nitrogen cycle.

Development of land-use regression models for exposure assessment to ultrafine particles in Rome, Italy

The health effects of long-term exposure to ultrafine particles (UFPs) are poorly understood. Data on spatial contrasts in ambient ultrafine particles (UFPs) concentrations are needed with fine resolution. This study aimed to assess the spatial variability of total particle number concentrations (PNC, a proxy for UFPs) in the city of Rome, Italy, using land use regression (LUR) models, and the correspondent exposure of population here living. PNC were measured using condensation particle counters at the building facade of 28 homes throughout the city. Three 7-day monitoring periods were carried out during cold, warm and intermediate seasons. Geographic Information System predictor variables, with buffers of varying size, were evaluated to model spatial variations of PNC. A stepwise forward selection procedure was used to develop a "base" linear regression model according to the European Study of Cohorts for Air Pollution Effects project methodology. Other variables were then included in more enhanced models and their capability of improving model performance was evaluated. Four LUR models were developed. Local variation in UFPs in the study area can be largely explained by the ratio of traffic intensity and distance to the nearest major road. The best model (adjusted R2 = 0.71; root mean square error = ±1,572 particles/cm³, leave one out cross validated R2 = 0.68) was achieved by regressing building and street configuration variables against residual from the "base" model, which added 3% more to the total variance explained. Urban green and population density in a 5,000 m buffer around each home were also relevant predictors. The spatial contrast in ambient PNC across the large conurbation of Rome, was successfully assessed. The average exposure of subjects living in the study area was 16,006 particles/cm³ (SD 2165 particles/cm³, range: 11,075–28,632 particles/cm³). A total of 203,886 subjects (16%) lives in Rome within 50 m from a high traffic road and they experience the highest exposure levels (18,229 particles/cm³). The results will be used to estimate the long-term health effects of ultrafine particle exposure of participants in Rome.

The characteristics changes of pH and EC of atmospheric precipitation and analysis on the source of acid rain in the source area of the Yangtze River from 2010 to 2015

Through the analysis of pH value, EC, precipitation and wind speed of 402 precipitation samples in the source region of the Yangtze River from January 1, 2010 to December 31, 2015, especially for the analysis of the 14 acid rain events. The results showed that: the acid rain in the source region of the Yangtze River was mainly affected by the southwest monsoon and the westerly circulation. The occurrence of acid rain mainly controlled by industrial pollution and other pollutants coming from India and other surrounding areas. And the other cause was that because of the Qinghai Tibet highway and the Qinghai Tibet railway, there were a lot of cars coming and going. And there were people in the summer to plateau tourism increased year by year, and more for self-driving travelling. This added additional pollutants (automobile exhaust) for the source of the Yangtze River. During the period of sampling, the variation range of pH value was from 4.0 to 8.57, with the mean was 6.37. And the range of EC was from 5.2 to 124.4 μs/cm, the average was 27.59 μs/cm. The order of conductivity in the four seasons was Spring > Winter > Summer > Autumn. And the order of pH in four seasons was Summer > Spring = Winter > Autumn. The results are also helpful for further understanding the acid rain in the Tibetan Plateau and providing scientific basis for the effective prevention and control of acid rain.

Dissolution and dissolution/permeation experiments for predicting systemic exposure following oral administration of the BCS class II drug clarithromycin

In order to save time and resources in early drug development, in vitro methods that correctly predict the formulation effect on oral drug absorption are necessary. The aim of this study was to 1) evaluate various BCS class II drug formulations with in vitro methods and in vivo in order to 2) determine which in vitro method best correlates with the in vivo results. Clarithromycin served as model compound in formulations with different particle sizes and content of excipients. The performed in vitro experiments were dissolution and dissolution/permeation experiments across two types of membrane, Caco-2 cells and excised rat intestinal sheets. The in vivo study was performed in rats. The oral absorption was enhanced by downsizing drug particles and by increasing the excipient concentration. This correlated strongly with the flux across Caco-2 cells but not with the other in vitro experiments. The insufficient correlation with the dissolution experiments can be partly explained by excipient caused problems during the filtration step. The very poor correlation of the in vivo data with the flux across excised rat intestinal sheets might be due to an artificially enlarged mucus layer ex vivo. In conclusion, downsizing BCS class II drug particles and the addition of surfactants enhanced the in vivo absorption, which was best depicted by dissolution/permeation experiments across Caco-2 cells. This setup is proposed as best model to predict the in vivo formulation effect. Also, this is the first study to evaluate the impact of the nature of the permeation membrane in dissolution/permeation experiments.

The anti-inflammatory and analgesic activity was tested in mice (Hot plate test) and rats (Carrageenan-induced paw edema test) respectively and their activity was compared to an aqueous solution of LC salt.ResultsThe aqueous solution showed a faster and shorter response whereas the optimised microemulsion increased significantly (p<0.01) the potency and duration of the analgesic and anti-inflammatory activity after deep intramuscular injection. The droplet size and the viscosity were key factors to control the drug release from the systems and enhance the effect of the formulations.ConclusionsThe microemulsion consisting of Labrafil®/Lauroglycol®/Polysorbate 80/water with LC (56.25/18.75/15/10, w/w) could be a promising formulation after buccal surgery due to its ability to control the drug release and significantly achieve greater analgesic and anti-inflammatory effect over 24h.

CD44+ cancer cell-induced metastasis: A feasible neck metastasis model

Neck metastasis of oral squamous cell carcinoma (OSCC) indicates a poor prognosis. Few neck metastasis models are currently available for drug trials and clinical research on OSCC. This study constructed a feasible animal model of neck metastasis by using CD44+ stem cell-like cancer cells. Real-time PCR was used to determine the expression levels of several reported cancer stem cell (CSC) markers, including CD44, CD133, ALDH1 and OCT4, in three OSCC cell lines. Magnetic sorting and DiO/DiD labelling were used to isolate subpopulations of cells and monitor cancer cell migration. Sorted and labelled CSC-like cells were injected into the tongue of nude mice. Tumour and metastatic lymph nodes were histologically examined through immunostaining and tracer staining. High rate of metastasis was observed in neck lymph nodes in CD44+ group. This phenomenon was confirmed in clinical OSCC patients, and CD44 expression levels were higher in tumours with lymph node metastasis than in carcinomas in situ. Therefore, we have successfully constructed a neck metastasis model by using CD44+ CSCs. This model can form tumour and show metastases within a short period of time compared with other models; additionally, this model may be used in short-term pharmacological experiments involving animals and in basic clinical studies.

Proteomic, toxicological and immunogenic characterization of Mexican west-coast rattlesnake (Crotalus basiliscus) venom and its immunological relatedness with the venom of Central American rattlesnake (Crotalus simus)

The venom of the Mexican west-coast rattlesnake (Crotalus basiliscus) was characterized for its protein composition, toxicological profile and immunogenic properties. This venom is composed of 68% Zn2+-dependent metalloproteinases (SVMPs), 14% phospholipases A2 (PLA2s), 11% serine proteinases, 4% SVMPs-inhibitor tripeptides (SVMP-ITs), 2% bradykinin-potentiating peptides (BPPs), 0.6% cysteine-rich secretory proteins (CRISPs), and 0.2% l-amino acid oxidases (LAAOs). SVMPs present in the venom are responsible for azocasein hydrolysis and hemorrhagic activity, but their contribution to the lethal activity of the venom in mice is masked by the neurotoxic activity of PLA2s, which in addition are also responsible for myotoxic activity. Despite its relatively high content of serine proteinases, the venom of C. basiliscus did not exert in vitro coagulant or in vivo defibrinogenating activities. The ability of antivenoms raised against the venoms of C. basiliscus and C. simus (from Costa Rica) to neutralize homologous and heterologous venoms revealed antigenic similarities between toxins of both venoms. Preclinical evaluation of an antivenom produced by using the venom of C. basiliscus as immunogen demonstrated that it is able to neutralize not only the most relevant toxic activities of C. basiliscus venom, but also those exerted by Costa Rican C. simus venom, including coagulant and defibrinogenating activities.Biological significanceThe Central American rattlesnake (Crotalus simus) is widely distributed from Mexico to west central Costa Rica, and induces an important number of envenomations in this region. On the other hand, the immunogenic mixture used by Laboratorios de Biológicos y Reactivos de Mexico S.A. (Birmex) to produce the snake antivenom more frequently used in Mexico does not include the venom of C. simus. This immunogenic mixture is composed by the venoms of the Fer-de-lance (Bothrops asper) and the Mexican west-coast rattlesnake (Crotalus basiliscus). We studied the protein composition, toxicological profile and immunogenic properties of the venom of C. basiliscus, and evaluated the ability of the Birmex antivenom to neutralize the venom of C. basiliscus and whether it cross-neutralizes the venom of C. simus from Costa Rica. Using proteomics analysis, in combination with in vitro and mouse tests, we determined that the venom of C. basiliscus is mainly composed by SVMPs, which confer proteolytic and hemorrhagic activities to the venom. Other major components of the venom of C. basiliscus are PLA2s, which are responsible for the myotoxic activity and are the main contributors to the lethal activity. Non-clotting SVSPs correspond to 11% of the venom. Minor components include SVMP-ITs, BPPs, CRISPs and LAAOs, which have not been associated with toxicity. The antibodies induced in horses by the venom of C. basiliscus are able to neutralize not only the most relevant toxic activities of the homologous venom, but also those exerted by Costa Rican C. simus venom, including coagulant and defibrinogenating activities. Our preclinical evaluation suggests that Birmex antivenom can be used to treat envenomations by Costa Rican adult C. simus snakebites, despite this venom not being included in the immunizing mixture.

Seasonal heat stress affects adipose tissue proteome toward enrichment of the Nrf2-mediated oxidative stress response in late-pregnant dairy cows

Environmental heat stress and metabolic stress during transition from late gestation to lactation are main factors limiting production in dairy cattle, and there is a complex interaction between them. Many proteins expressed in adipose tissue are involved in metabolic responses to stress. We aimed to investigate the effects of seasonal heat stress on adipose proteome in late-pregnant cows, and to identify biomarkers of heat stress. Late pregnant cows during summer heat stress (S, n=18), or during the winter season (W, n=12) were used. Subcutaneous adipose tissue biopsies sampled 14days prepartum from S (n=10) and W (n=8) were analyzed by intensity-based, label-free, quantitative shotgun proteomics (nano-LC–MS/MS). Plasma concentrations of malondialdehyde and cortisol were higher in S than in W cows. Proteomic analysis revealed that 107/1495 proteins were differentially abundant in S compared to W (P<0.05 and fold change of at least ±1.5). Top canonical pathways in S vs. W adipose were Nrf2-mediated oxidative stress response, acute-phase response, and FXR/RXR and LXR/RXR activation. Novel biomarkers of heat stress in adipose tissue were found. These findings indicate that seasonal heat stress has a unique effect on adipose tissue in late-pregnant cows.SignificanceThis work shows that seasonal heat stress increases plasma concentrations of the oxidative stress marker malondialdehyde and cortisol in transition dairy cows. As many proteins expressed in the adipose tissue are involved in metabolic responses to stress, we investigated the effects of heat stress on the proteome of adipose tissue from late-pregnant cows during summer or winter seasons. We demonstrated that heat stress enriches several stress-related pathways, such as the Nrf2-mediated oxidative stress response and the acute-phase response in adipose tissues. Thus, environmental heat stress has a unique effect on adipose tissue in late-pregnant cows, as part of the regulatory adaptations to chronic heat load during the summer season. In addition, this study presents the widest available dataset of adipose tissue proteome in dairy cows, and revealed several novel biomarkers of heat stress in adipose tissue of dairy cows, the use of which awaits further validation.

Characterisation of the Bordetella pertussis secretome under different media

Our understanding of the Bordetella pertussis secretome remains limited including the role of different growth conditions in the secretome. In this study the secretome of L1423, a clinical isolate from the 2008–2012 Australian epidemic, cultured on Stainer-Scholte (SS) and Thalen-IJssel (THIJS) media for 12h was characterised using liquid chromatography-mass spectrometry (LC-MS/MS). In the supernatant, LC-MS/MS identified 260 proteins with 143 bioinformatically predicted to be secreted. Eighty percent of proteins were identified in both media. Proteins secreted were functionally associated with cell surface (41%), pathogenicity (16%) and transport (17%). The most abundant proteins identified were pathogenic proteins including toxins (PtxA and CyaA), adhesins (TcfA) and type III secretion (T3SS) proteins. There were 46 proteins found uniquely in THIJS including 8 virulence associated proteins. These included T3SS proteins, adhesins (FhaL and FhaS) and a putative toxin (BP1251). Nine proteins were found uniquely in SS and these were metabolic and transport-related proteins. None of the unique proteins detected in SS were known to be virulence associated. This study found that THIJS promotes secretion of virulence factors based on the number of unique virulence proteins found and may be a growth media of choice for the study of B. pertussis virulence and vaccine development.Biological significanceOver the past two decades, the number of B. pertussis notifications has risen despite vaccination. There is a greater need to understand the biology behind B. pertussis infections. The secretome of B. pertussis in two different media was characterised using LC-MS/MS. The results show that THIJS promotes secretion of importance virulence factors which may be important for the development of vaccines.

Analysis of the herbicidal mechanism of 4-hydroxy-3-methoxy cinnamic acid ethyl ester using iTRAQ and real-time PCR

Absolute quantitation (iTRAQ) is the latest development in the new quantitative proteomics technology for high-throughput identification and quantitation of proteins. The mechanisms underlying the 4-hydroxy-3-methoxy cinnamic acid ethyl ester treatment in Arabidopsis thaliana was investigated. Deficiency-induced changes in the protein profile of A. thaliana caused by this compound were analyzed using iTRAQ and quantitative real-time PCR. A total of 2909 proteins were quantified, of which 49 and 34 proteins were upregulated and downregulated, respectively, in the experimental plants compared with the controls. Treatment results showed that numerous proteins were involved in photosystemII, energy metabolism, and cell structure formation. Based on the upregulated and downregulated proteins, high amount of AT4G21280 protein acted on the oxygen-evolving enhancer protein 3-1, while low amount of AT1G10340 protein affected the catabolic process of the photosystemII-associated light-harvesting complex II. We selected these proteins to preliminarily verify the expression of proteins using quantitative real-time PCR to provide a reliable basis for further studies after proteomics analysis. Results show that the combined use of iTRAQ and quantitative real-time PCR provides an effective method to study proteins, leading to the determination of a new herbicide mechanism.

Positive relationship between subsequent chemotherapy and overall survival in pancreatic cancer: meta-analysis of postprogression survival for first-line chemotherapy

Abstract

Purpose

To gain a better understanding of the impact of postprogression survival (PPS) and post-trial anticancer therapy on overall survival (OS) in first-line pancreatic cancer patients.

Methods

A literature search identified 54 randomized trials, focusing on gemcitabine monotherapy to eliminate effects of heterogeneity of first-line regimens. We evaluated the relation between OS and either progression-free survival (PFS) or PPS. We also examined whether any association might be affected by the year of completion of trial enrollment.

Results

For all 54 trials, PPS was strongly associated with OS (r = 0.844), whereas PFS was moderately associated with OS (r = 0.623). Average OS and PPS were significantly longer in recent trials than in older trials, (7.29 versus 6.15 months, p < 0.001) and (3.64 versus 2.86 months, p < 0.001), respectively. The correlation between OS and PPS in recent trials was much stronger than that in older trials (r = 0.846 versus 0.729). The relation between OS and PFS in recent and older trials did not differ (r = 0.595 versus 0.563). The percentage of patients with post-trial treatment was significantly higher in recent trials than in older trials (52.7 versus 39.7%, p < 0.001). The rate of post-trial anticancer therapy was significantly associated with OS (r = 0.910).

Conclusions

We found an increase in median PPS in accordance with an increase in median OS in recent trials compared with older trials and that rate of post-trial anticancer therapy was strongly associated with median OS. It is important that researchers be aware of these findings in designing clinical trials of first-line chemotherapy for pancreatic cancer patients.

Exendin-4 inhibits structural remodeling and improves Ca2+ homeostasis in rats with heart failure via the GLP-1 receptor through the eNOS/cGMP/PKG pathway

The glucagon-like peptide-1 receptor (GLP-1R) agonist exendin-4 is a long-acting analogue of GLP-1, which stimulates insulin secretion and is clinically used in the treatment of type 2 diabetes. Previous studies have demonstrated that GLP-1 agonists and analogues serve as cardioprotective factors in various conditions. Disturbances in calcium cycling are characteristic of heart failure (HF); therefore the aim of this study was to investigate the effect of exendin-4 (a GLP-1 mimetic) on the regulation of calcium handling and identify the underlying mechanisms in a heart failure (HF) rat model after myocardial infarction (MI). Rats underwent surgical ligation of the left anterior descending coronary artery or sham surgery prior to infusion with vehicle, exendin-4, or exendin-4 and exendin9-39 for four weeks. Exendin-4 treatment decreased MI size, suppressed chamber dilation, myocyte hypertrophy and fibrosis, and improved in vivo heart function in the rats subjected to MI. Exendin-4 resulted in an increase in circulating GLP-1, and GLP-1R in ventricular tissues. Additionally, exendin-4 activated the eNOS/cGMP/PKG signaling pathway and inhibited the Ca2+/calmodulin-dependent kinase II (CaMKII) pathways. Myocytes isolated from exendin-4-treated hearts displayed higher Ca2+ transients, higher sarcoplasmic reticulum Ca2+ content and higher L-type Ca2+ current densities than MI hearts. Exendin-4 treatment restored the protein expression of sarcoplasmic reticulum Ca2+ uptake ATPase (SERCA2a), phosphorylated phospholamban (PLB) and Cav1.2 and decreased the levels of phosphorylated ryanodine receptor (RyR). Moreover, the favorable effects of exendin-4 were significant inhibited by exendin9-39 (a GLP-1 receptor antagonist). Exendin-4 treatment of a HF rat model after MI inhibited cardiac and cardiomyocytes progressive remodeling. In addition, Ca2+ handling and its molecular modulation were also improved by exendin-4 treatment. The beneficial effects of exendin-4 on cardiac remodeling may be mediated through activation of the eNOS/cGMP/PKG pathway.

Gastric bypass in the pig increases GIP levels and decreases active GLP-1 levels

Gastric bypass surgery results in remission of type 2 diabetes in the majority of patients. The incretin hormones glucagon-like peptide 1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) have been implicated in the observed remission. Most knowledge so far has been generated in obese subjects. To isolate the surgical effects of gastric bypass on metabolism and hormone responses from the confounding influence of obesity, T2D or food intake, we performed gastric bypass in lean pigs, using sham-operated and pair-fed pigs as controls. Thus, pigs were subjected to Roux-en-Y gastric bypass (RYGB) or sham-surgery and oral glucose tolerance tests (OGTT). RYGB- and sham-pigs exhibited similar basal and 120-minute glucose levels in response to the OGTT. However, RYGB-pigs had approximately 1.6-fold higher 30-min glucose (p<0.01). Early insulin release (EIR) was enhanced approximately 3.5-fold in the RYGB-pigs (p<0.01). Furthermore, GIP release, both the acute and sustained release (p<0.001 and p<0.01, respectively) were increased approximately 2.5-fold and 1.4-fold, respectively, in RYGB-pigs. While total GLP-1 release increased approximately 2.1-fold after RYGB (p<0.001), active GLP-1 was 33% lower (p<0.01). Interestingly basal DPP4-activity was approximately 3.2-fold higher in RYGB-pigs (p<0.001).In conclusion, RYGB in lean pigs increases the response of GIP, total GLP-1 and insulin, but reduces levels of active GLP-1 in response to an oral glucose load. These data challenge the role of active GLP-1 as a contributor to remission from diabetes after RYGB.
