The earliest occurrence of a newly described domesticate in Eastern North America: Adena/Hopewell communities and agricultural innovation

S02784165.gif

Publication date: March 2018
Source:Journal of Anthropological Archaeology, Volume 49
Author(s): Natalie G. Mueller
Archaic and Woodland period communities in eastern North America domesticated a suite of annual seed crops referred to the Eastern Agricultural Complex (EAC), some of which subsequently fell out of cultivation and were lost. Recently, a domesticated sub-species of one of these lost crops, erect knotweed (Polygonum erectum) has been described. This paper reports the earliest example of this domesticated sub-species, which was recovered from a sub-mound context at an Adena/Hopewell site in central Kentucky (Walker-Noe, 15Gd56) dating to c. 1 AD. Contemporary Middle Woodland erect knotweed assemblages from habitation sites in western Illinois are not domesticated. A review of the paleoethnobotanical record suggests that farmers on the western front of the Appalachian Mountains developed several innovative agricultural practices, beginning around 1000 BC, that subsequently were adopted across the core area of EAC cultivation. The ethnography and sociology of 20th and 21st century farmer networks suggests that Adena/Hopewell exchange and community integration at mounds and earthworks may have been instrumental to this process. Additional analyses of botanical assemblages from mounds and earthworks, especially morphometric analyses of crop seeds, are necessary to test this hypothesis. The dynamics of social learning involved in this process may also be implicated in the spread of crop varieties and agricultural techniques in other regions.

Proteomics reveals key proteins participating in growth difference between fall dormant and non-dormant alfalfa in terminal buds

Publication date: 20 February 2018
Source:Journal of Proteomics, Volume 173
Author(s): Hongqi Du, Yinghua Shi, Defeng Li, Wenna Fan, Yanhua Wang, Guoqiang Wang, Chengzhang Wang
To explore the molecular mechanism of growth differences between fall dormant (FD) and non-FD alfalfa, we conducted iTRAQ-based quantitative proteomics on terminal buds of Maverick (FD) and Cuf101 (non-FD) cultivars, identified differential abundance protein species (DAPS) and verified expression profiling of certain corresponding mRNA by qRT-PCR. A total of 3872 protein species were annotated. Of the 90 DAPS, 56 and 34 were respectively up- and down-accumulated in Maverick, compared to Cuf101. They were grouped into 35 functional categories and enriched in seven pathways. Of which, auxin polar transport was up-regulated, while phenylpropanoid biosynthesis, pyruvate metabolism and transportation, vitamin B1 synthesis process and flavonoid biosynthesis were down-regulated in Maverick, comparing with Cuf101. In Maverick, mRNA abundances of l-asparaginase, chalcone and stilbene synthase family protein, cinnamyl alcohol dehydrogenase-like protein, thiazole biosynthetic enzyme, pyruvate dehydrogenase E1 beta subunit, and aldo/keto reductase family oxidoreductase were significantly lower at FD than at other stages, and lower than in Cuf101. We also observed opposite mRNA profiles of thiazole biosynthetic enzyme, chalcone and stilbene synthase family protein, pyruvate dehydrogenase E1 beta subunit in both cultivars from summer to autumn. Our results suggest that these DAPS could play important roles in growth difference between FD and non-FD alfalfa.Biological significanceUp to now, as far as we know, currently the proteins related with the growth differences between FD and non-FD alfalfa cultivars in autumn have not yet been identified in terminal buds. This study identified the protein species expressed in alfalfa terminal buds, selected differentially abundant protein species in terminal buds between Maverick (FD) and Cuf101 (non-FD) cultivars in autumn and identified the important protein species participated in the growth differences. This study lays a foundation for further investigation of the molecular mechanism of the growth differences between FD and non-FD alfalfa and the cultivation of advanced alfalfa cultivars.

Revealing critical mechanisms of BR-mediated apple nursery tree growth using iTRAQ-based proteomic analysis

Publication date: 20 February 2018
Source:Journal of Proteomics, Volume 173
Author(s): Liwei Zheng, Juanjuan Ma, Lizhi Zhang, Cai Gao, Dong Zhang, Caiping Zhao, Mingyu Han
Brassinosteroid is identified as an important hormone. However, information about brassinosteroid has not been fully elucidated, and few studies concerned its role in apple. The aim of this work was to study the role of brassinosteroid for apple tree growth. In our study, the effect of brassinosteroid on apple nursery tree was analyzed. The biomass, cell size and xylem content of apple nursery tree were obviously evaluated by brassinosteroid treatment; mineral elements contents, photosynthesis indexes, carbohydrate level and hormone contents were significantly high in brassinosteroid treated trees. To explore the molecular mechanisms of these phenotypic differences, iTRAQ-based quantitative proteomics were used to identify the expression profiles of proteins in apple nursery tree shoot tips in response to brassinosteroid at a key period (14days after brassinosteroid treatment). A total of 175 differentially expressed proteins were identified. They were mainly involved in chlorophyII biosynthesis, photosynthesis, carbohydrate metabolism, glycolysis, citric acid cycle, respiratory action, hormone signal, cell growth and ligin metabolism. The findings in this study indicate that brassinosteroid mediating apple nursery tree growth may be mainly through energy metabolism. Important biological processes identified here can be useful theoretical basis and provide new insights into the molecular mechanisms of brassinosteroid.Biological significanceBrassinosteroid is very important for plant growth and development. However, the molecular mechanism of brassinosteroid mediating growth process is not perfectly clear in plant, especially in apple nursery tree. We used a combination of physiological and bioinformatics analysis to investigate the effects of brassinosteroid on apple nursery tree growth and development. The data reported here demonstrated that brassinosteroid regulates apple nursery tree growth mainly through energy metabolism. Therefore it can provide a theoretical basis from energy points for developing dwarfed or compact apple trees. This will benefit for low orchard management cost as well as early bearing, and high fruit yield as well as quality.

Manufacturing of an electrochemical biosensing platform based on hybrid DNA hydrogel: Taking lung cancer-specific miR-21 as an example

Publication date: 30 April 2018
Source:Biosensors and Bioelectronics, Volume 103
Author(s): Shuopeng Liu, Wenqiong Su, Yulong Li, Lulu Zhang, Xianting Ding
DNA hydrogel garnered increasing attention in the sensing and medical field owing to its native biocompatibility and mechanical stability. While electrochemistry serves as a quantitative and sensitive detection technique, electrochemical DNA hydrogel biosensor is rarely reported. Here, for the first time, we report an electrochemical biosensor based on hybrid DNA hydrogel immobilized on indium tin oxide/polyethylene terephthalate (ITO/PET) electrode for the detection of lung cancer-specific microRNA, miR-21. The biosensor is capable of detecting miR-21 at a concentration as low as 5nM (1 pmol) and linear read-out from 10nM to 50μM. Ferrocene-tagged recognition probes were cross-linked with DNAs grafted on the polyacrylamide backbones to form the hybrid DNA hydrogel, which was further immobilized on 3-(trimethoxysilyl)propyl methacrylate (KH 570) treated ITO electrode. When the recognition probe was hybridized with the target miR-21, the hydrogel dissolved, producing a loss of ferrocene tags and a reduction in current, detected by Cyclic Voltammetry (CV) and Differential Pulse Voltammetry (DPV). The material characteristics of the biosensor were verified using contact angle meter and Energy Dispersive Spectrometer (EDS). This novel biosensor holds great promise in early sensitive clinical diagnosis for a variety of cancer-specific biomarkers due to the flexible sequence design of the recognition probe.

Role of Physical Activity and Exercise in Alleviating Cognitive Impairment in People With Epilepsy

alertIcon.gif

Publication date: Available online 21 December 2017
Source:Clinical Therapeutics
Author(s): Jane B. Allendorfer, Ricardo M. Arida
Many persons with epilepsy (PWE) experience problems with a wide range of cognitive functions, including learning, memory, attention, and executive control. These deficits in cognition result in diminished quality of life for PWE and are related to many factors, including the etiology of their epilepsy, recurrent seizures, side effects of antiseizure medications, or a combination of these factors. Various treatments to ameliorate cognitive deficits experienced by PWE have been implemented, although noninvasive and nonpharmacologic strategies may be more appealing options due to their relatively low cost, reduced risk of side effects, and/or reduced potential interactions with antiseizure medications. Physical activity and exercise may improve cognition in PWE but have not been well researched in this respect. To date only 1 study has directly investigated the effects of exercise on cognition in PWE, and it showed improved performance on tests of attention and executive function. The goal of the present article was to examine how increased physical activity and exercise contributes to 3 strategies (reducing seizure frequency, reducing epileptiform discharges, and decreasing symptoms of depression) that have been described as having a positive impact on cognition in PWE, as well as highlight related findings in experimental models of epilepsy. There is a definite need for more randomized controlled trials to establish greater clinical evidence for the use of physical activity and exercise in ameliorating cognitive impairment in PWE. We also need to better understand the factors contributing to reduced physical activity in PWE, as well as ways to overcome such barriers. With the available research in the area of exercise in epilepsy showing positive results, and a supportive research climate encouraging PWE to engage in greater physical activity overall, further investigations into the relationships between physical activity and cognition in epilepsy are warranted.

PurposeThe efficacy and tolerability of vitamin K antagonists (VKAs) depends on the quality of anticoagulant control, reflected by the mean time in therapeutic range (TTR) of international normalized ratio 2.0 to 3.0. In the present study, we aimed to investigate the association between TTR and change in TTR (ΔTTR) with the risk of mortality and clinically significant events in a consecutive cohort of atrial fibrillation (AF) patients.MethodsWe included 1361 AF patients stable on VKAs (international normalized ratio 2.0−3.0) during at least the previous 6 months. After 6 months of follow-up we recalculated TTR, calculated ΔTTR (ie, the difference between baseline and 6-month TTRs) and investigated the association of both with the risk of mortality and "clinically significant events" (defined as the composite of stroke or systemic embolism, major bleeding, acute coronary syndrome, acute heart failure, and all-cause deaths).FindingsThe median ΔTTR at 6 months of entry was 20% (interquartile range 0−34%), 796 (58.5%) patients had a TTR reduction of at least 20%, while 330 (24.2%) had a TTR <65%. During follow-up, 34 (2.5% [4.16% per year]) patients died and 61 (4.5% [7.47% per year]) had a clinically significant event. Median ΔTTR was significantly higher in patients who died (35.5% vs 20%; P = 0.002) or sustained clinically significant events (28% vs 20%; P = 0.022). Based on Cox regression analyses, the overall risk of mortality at 6 months for each decrease point in TTR was 1.02 (95% CI, 1.01−1.04; P = 0.003), and the risk of clinically significant events was 1.01 (95% CI, 1.00−1.03; P = 0.028). Patients with TTR <65% at 6 months had higher risk of mortality (hazard ratio = 2.96; 95% CI, 1.51−5.81; P = 0.002) and clinically significant events (hazard ratio = 1.71; 95% CI, 1.01−2.88; P = 0.046).ImplicationsOur findings suggest that in AF patients anticoagulated with VKAs, a change in TTR over 6 months (ie, ΔTTR) is an independent risk factor for mortality and clinically significant events. Even in a cohort with good anticoagulation control, the risk for mortality and clinically significant events increases with every point deterioration of TTR.

Influence of low molecular weight compounds associated to cashew (Anacardium occidentale L.) fiber on lipid metabolism, glycemia and insulinemia of normal mice

Publication date: Available online 21 December 2017

The objective of this study was to evaluate the effect of cashew fiber wheter or not associated with low molecular weight compounds on lipid profile, abdominal adiposity, glycemia and insulinemia and serum concentration of ghrelin and leptin hormones from normal mice. We evaluated the cashew fiber in its integral form (IcF) and the cashew fiber after being submitted to the process of extraction of low molecular weight metabolites (cFSM). The two fibers were incorporated into the feed of male Swiss mice for 15 weeks. At the end of the treatment, blood glucose, total cholesterol and fractions, triglycerides, urea, AST, ALT, hormone levels of insulin, leptin and ghrelin were analyzed, as well as GSH and hepatic MDA. The consumption of cFSM diet promoted reduction of glycemia, insulin and ghrelin. Animals fed the IcF diet showed hyperlipidemia, hyperleptinemia and increased abdominal fat. In addition, no changes were observed in the lipid profile of the animals, in the abdominal adiposity and in the leptin hormone in the group that received cFSM diet. We conclude that the removal of small molecules is fundamental to use cashew fiber as a functional food with possible health benefits.

Extracellular ATP signaling and clinical relevance

S15216616.gif

Publication date: Available online 20 December 2017
Source:Clinical Immunology
Author(s): Lei Dou, Yi-Fa Chen, Peter J. Cowan, Xiao-ping Chen
Since purinergic signaling was discovered in the early 1970s, it has been shown that extracellular nucleotides, and their derivative nucleosides, are released in a regulated or unregulated manner by cells in various challenging settings and then bind defined purinergic receptors to activate intricate signaling networks. Extracellular ATP plays a role based on different P2 receptor subtypes expressed on specific cell types. Sequential hydrolysis of extracellular ATP catalyzed by ectonucleotidases (e.g. CD39, CD73) is the main pathway for the generation of adenosine, which in turn activates P1 receptors. Many studies have demonstrated that extracellular ATP signaling functions as an important dynamic regulatory pathway to coordinate appropriate immune responses in various pathological processes, including intracellular infection, host-tumor interaction, pro-inflammation vascular injury, and transplant immunity. ATP receptors and CD39 also participate in related clinical settings. Here, we review the latest research in to the development of promising clinical treatment strategies.
