Is Human Sebum the Source of Skin Follicular Ultraviolet-Induced Red Fluorescence? A Cellular to Histological Study

Background: The ultraviolet-induced red fluorescence (UVRF) from human skin follicles was suggested to be a result of Propionibacterium acnes and was used for the monitoring of acne. More recent studies suggested that the UVRF may be more related to sebum rather than to microorganisms. Objective: To clarify whether human sebum or follicular microorganisms are the source of UVRF. Methods: We examined the fluorescence of human-derived SZ95 sebocytes, human sebaceous glands, sebum extracted from the sebaceous glands, and bacteria isolated from human hair follicles under ultraviolet light. Results: SZ95 sebocytes, human sebaceous glands, and sebum do not emit UVRF. Two types of UVRF peaking at about 635 nm and at about 620 nm were detected in P. acnes and Staphylococcus epidermidis, respectively. This is the first report that S. epidermidis emits UVRF when it is anaerobically cultured and then exposed to air. Conclusion: Human follicular UVRF is emitted by resident bacteria, not by sebum. Therefore, UVRF may be used to monitor certain species of skin microorganisms.

Presence of artificial radionuclides in samples from potable water and wastewater treatment plants

Human activity, such as the operation of nuclear power plants (NPPs) and the use of radionuclides in nuclear medicine, results in the presence of artificial radionuclides in surface waters, which may even reach potable water treatment plants (PWTPs) and wastewater treatment plants (WWTPs).In this study, water and sludge samples from a PWTP are radiologically monitored. The incoming water of the plant is influenced by the presence of an NPP upstream. Two WWTPs receiving wastewater from medical centres and other origins are also studied. As a result, 131I, 60Co and 137Cs have been determined in the dewatered sludge samples from the PWTP, while 131I, 99mTc, 67Ga and 111In were detected in the sludge samples from the WWTPs. The radionuclide activities in the influent water from the WWTPs studied were lower than the minimum detectable activity values. Therefore, on the basis of our results, the analysis of sludge samples is very useful as it enables the concentration of any radionuclides that may be present in the incoming water.Lastly, as higher activity of 131I was detected in the samples studied, the total effective dose was assessed for WWTP workers, as they handle dewatered sludge containing this radionuclide. It can be concluded that there is no risk in terms of total exposure.

Animal abuse as an outcome of poor emotion regulation: A preliminary conceptualization

Animal abuse is an under-reported yet prevalent form of both passive and active forms of aggressive behavior. Its severe and upsetting consequences are not only experienced by the victims themselves, but also others in proximity (e.g., pet owners). Despite this, research and theory focusing on the motivations for such behavior appear to be sparse and limited in development when compared to other types of offending behavior, such as interpersonal violence. This article examines the motivations that underlie animal abuse and the maladaptive emotion regulation techniques that facilitate this type of behavior. We focus on two specific emotion regulation styles that have been implicated in existing literature; that is, the mis-regulation and under-regulation of emotions. Based on existing research and theories, we posit that the facilitative role emotion regulation plays in the perpetration of animal abuse is vital in our understanding of how and why this abuse occurs. In this article, we present a preliminary conceptualization of animal abuse behavior that depicts emotion regulation as a pivotal factor in key explanatory pathways.

Addressing violence against educators through measurement and research

Violence against educators is a significant understudied phenomenon that has been largely ignored by policy makers and funders. The primary goal of this paper is to advance measurement and research on educator safety and victimization. We conducted a comprehensive systematic review of the extant literature (1988 to 2016). Although the number of studies has increased dramatically over the past decade, this review revealed only 37 studies in 28 journals (49,581 educators and 91,099 students). On the basis of this review, we highlight key findings, propose a conceptual framework for measurement development, outline key variables for the design of educator safety registries, and identify research priorities. Multidimensional teacher safety assessment systems and registries for educator safety and victimization are crucial to the advancement of the field and the safety of our schools.

South Africa has instituted many legislative and constitutional changes since 1994 to combat violence. Regardless of these legislative developments, South Africa has some of the most endemic rates of violence in the world with most of these high rates of violence located within family violence. In response, South African legislation drew from a number of international and regional commitments as well as constitutional amendments to facilitate an enhanced family environment which promoted family well-being and functioning. However, legislatively, family violence is still fragmented in its respective subsets of intimate partner violence, child abuse and elder abuse. Disregarding family violence in the holistic sense, fragmented sectors and efforts to comprehensively and adequately prevent or reduce the rates of family violence. Through a family centered perspective, this paper aimed to review the various legislation used to address family violence within the South African context.

Evaluation on Heterogeneity of Fatty Liver in Rats: A Multiparameter Quantitative Analysis by Dual Energy CT

Rationale and ObjectivesTo investigate the value of using rapid kVp switching dual energy computed tomography (rsDECT) for the multi-parameter analysis of the heterogeneity of fatty liver in rat.Materials and MethodsTwenty-four male rats were randomly divided into experimental (n = 16) and control (n = 8) groups. Four rats in the experimental group and two in the control group were examined by rsDECT every 3 weeks starting from week 8. The liver fat contents (LFC) of the left and right liver lobes were measured on the fat(water)-based material decomposition images to calculate fat content and CT value of liver and spleen were measured on the 70keV monochromatic images to calculate the liver-to-spleen CT value ratio [(L/S)70 keV] and difference at 70keV[(L−S)70 keV], and the spectral curve slopes of the left and right liver lobes (Slope-L, Slope-R). Measurements were evaluated statistically.ResultsThe statistical analysis showed that the (L/S)70 keV, (L−S)70 keV, Slope and LFC in the different fatty liver groups were all significantly different (p < 0.05). Pearson correlation analysis results showed that the rsDECT results of the left and right liver lobes correlated with the fat percentage from pathological analysis (p < 0.05), with the left liver lobe having better correlation. Paired t-tests showed that the fat percentage of the left liver lobe was significantly higher than that of the right one, while (L/S)70 keV, and (L−S)70 keV were significantly lower. Diagnostic analysis using ROC curve showed that the areas under the curves with parameters of the left liver lobe were also greater than those of the right liver lobe.ConclusionrsDECT multi-parameter imaging could quantitatively evaluate the heterogeneity of fat deposition in the liver, providing valuable information for the accurate and effective assessment of the heterogeneity of fatty liver.

Immune checkpoint blockade therapy for cancer: An overview of FDA-approved immune checkpoint inhibitors

Although T lymphocytes have long been appreciated for their role in the immunosurveillance of cancer, it has been the realization that cancer cells may ultimately escape a response from tumor-reactive T cells that has ignited efforts to enhance the efficacy of anti-tumor immune responses. Recent advances in our understanding of T cell immunobiology have been particularly instrumental in informing therapeutic strategies to overcome mechanisms of tumor immune escape, and immune checkpoint blockade has emerged as one of the most promising therapeutic options for patients in the history of cancer treatment. Designed to interfere with inhibitory pathways that naturally constrain T cell reactivity, immune checkpoint blockade releases inherent limits on the activation and maintenance of T cell effector function. In the context of cancer, where negative T cell regulatory pathways are often overactive, immune checkpoint blockade has proven to be an effective strategy for enhancing the effector activity and clinical impact of anti-tumor T cells. Checkpoint inhibitors targeting CTLA-4, PD-1, and PD-L1 have yielded unprecedented and durable responses in a significant percentage of cancer patients in recent years, leading to U.S. FDA approval of six checkpoint inhibitors for numerous cancer indications since 2011. In this review, we highlight the clinical success of these FDA-approved immune checkpoint inhibitors and discuss current challenges and future strategies that must be considered going forward to maximize the efficacy of immune checkpoint blockade therapy for cancer.

Application of Box–Behnken design and desirability function in the optimization of Cd(II) removal from aqueous solution using poly(o-phenylenediamine)/hydrous zirconium oxide composite: equilibrium modeling, kinetic and thermodynamic studies

Abstract

In this research work, poly(o-phenylenediamine) was incorporated into the hydrous zirconium oxide matrix to form poly(o-phenylenediamine)/hydrous zirconium oxide composite which is used for the removal of Cd(II) from aqueous solution. The characterization of the material was done based on FTIR, XRD, SEM, and TGA-DTA. The effects of contact time, pH, adsorbent dose, and initial concentration of Cd(II) on the removal of Cd(II) were studied by performing 29 sets of sorption runs using Box–Behnken design combined with response surface methodology (RSM). Various isotherm models were tested to describe the adsorption equilibrium. The adsorption equilibrium data fitted well with Freundlich isotherm model. The maximum adsorption capacity of 66.66 mg g−1 was obtained from Langmuir isotherm. The pseudo-second-order kinetic model described the adsorption kinetics more accurately. Diffusion-based kinetics such as intraparticle diffusion and Bangham's model suggested that both film and intraparticle pore diffusion were involved in the adsorption process. The Elovich model pointed towards the chemisorption. The investigation of desorption and regeneration suggested that the material can be used as an effective sorbent for removal of Cd(II) from aqueous system.

Mutations of canonical Wnt signaling pathway genes frequently occur in cancer and lead to abnormal accumulation of the key effector β-catenin. Over the past decades, a number of Wnt inhibitors have been identified through high-throughput screenings, however, very few of them target β-catenin directly, raising questions regarding its druggability. Here, we review Wnt inhibitors with a focus on small molecules that directly bind β-catenin, discuss the druggability of β-catenin, and why it has rarely been targeted, especially in the cellular context. We also propose strategies to develop small molecule binding and depleting cellular β-catenin, which are generally applicable to other difficult-to-drug or yet-to-be-drugged targets.

Microbial fuel cell and membrane bioreactor coupling system: recent trends

Abstract

Membrane bioreactor (MBR) and microbial fuel cell (MFC) are new technologies based on microbial process. MBR takes separation process as the core to achieve the high efficient separation and enrichment the beneficiation of microbes during the biological treatment. MFC is a novel technology based on electrochemical process to realize the mutual conversion between biomass energy and electric energy, in order to solve the problems of serious membrane fouling and low efficiency of denitrification in membrane bioreactor, the low power generation efficiency, and unavailability of bioelectric energy of MFC. In recent years, MFC-MBR coupling system emerged. It can effectively mitigate the membrane fouling and reduce the excess sludge production. Simultaneously, the electricity can be used effectively. The new coupling system has good prospects for development. In this paper, we summarized the research progresses of the two kinds of coupling systems in recent years and analyzed the coupling structure and forms. Based on the above, the future development fields of the MFC-MBR coupling system were prospected.

Patterns of complications following urinary tract reconstruction after multivisceral surgery in colorectal and anal cancer

BackgroundMulti-visceral resection, including parts of the urinary tract, is sometimes warranted to achieve cancer clear resection margins and optimize survival in patients with locally advanced colorectal and anal cancer. The aim of this study was to assess morbidity after urinary tract reconstruction dictated by colorectal and anal malignancy and to identify potential predictors of urological complications.MethodsAll patients undergoing surgery for colorectal or anal malignancy, including urinary tract resection and synchronous reconstruction, performed at the Karolinska University Hospital during 2004-2015 were included in this retrospective cohort study. Data was collected from medical records with follow-up until at least one year after the index surgery. Complications were graded according to the Clavien-Dindo classification system of surgical complications.ResultsThe study included 189 patients; 121 underwent cystectomy and 68 partial ureter resection. The rate of high grade urological complications was 22%. The risk of major urological complications was significantly higher in patients subjected to ureter resection compared to after cystectomy (OR 2.60, 95% CI 1.23-5.49). Also, preoperative radiotherapy and intestinal anastomotic dehiscence significantly increased the risk of high grade urological complications.ConclusionTo achieve potentially curative resections with uninvolved margins in patients with locally advanced colorectal and anal cancer, multi-visceral resection including urinary tract reconstruction can be performed with reasonable morbidity.

Fate of PAHs in the vicinity of aluminum smelter

Abstract

Investigation has been carried out in the vicinity of an aluminum smelter located in the industrialized town of Konin. Concentrations of 14 polycyclic aromatic hydrocarbons (PAHs) were determined in grass, spruce needles, and soil collected in the period of the smelter operation and several years after its closing. Significant changes in the quantity of PAHs and their profiles observed in the two measuring periods, stressing the importance of aluminum production with regard to PAH emission. It was confirmed by very high values of the carcinogenic potential (CP) found for PAHs accumulated in grass and soil when compared to the values found in urban and remote sites. PAH ratio rates used as a tool for identifying emission sources showed a pyrogenic origin of PAHs in both periods; the ratios in the period of the smelter running activity were similar to those found in other studies carried out near aluminum smelters. Grass turned out to be a good biomonitor of PAHs similarly to commonly used leaves of various tree species. The use of four age classes of spruce needles, some of which were subjected to emission from the smelter, showed that such approach could serve as an analysis tool for describing retrospective pollution.

Evaluation of psychiatric hospital wastewater toxicity: what is its impact on aquatic organisms?

Abstract

The primary source of pharmaceuticals to the aquatic environment is the discharge of wastewater effluents. Pharmaceuticals are a large and diverse group of compounds. Among them, psychotropic substances are particularly interesting to study due to their specific known mode of action. The present study was performed to investigate the effects of wastewater effluents from a psychiatric hospital wastewater treatment plant (WWTP) on several aquatic organisms. All the analyzed pharmaceuticals (10 compounds) were detected in WWTP effluents as well as in the receiving river. Although the environmental concentrations were generally at trace levels (ng L−1 to μg L−1), induce toxic effects were observed. This study showed the effects of the WWTP effluents on the oogenesis and/or embryogenesis of amphipod crustacean Gammarus fossarum, Japanese fish medaka Oryzias latipes, mollusk Radix peregra, and planarian Schmidtea polychroa. A decrease of the number of oocytes and produced embryos was observed for G. fossarum and S. polychroa. Similarly, the hatching rate of R. peregra was affected by effluents. In the receiving river, the macroinvertebrate community was affected by the wastewater effluents discharge.

Application of the bespoke solid-phase extraction protocol for extraction of physiologically-active compounds from vegetable oils

A possibility of harvesting the physiologically-active compounds from the vegetable oils using bespoke resin and optimised solid-phase extraction (SPE) purification method is demonstrated. The paper describes the application of SPE protocol, which was originally developed, using sunflower oil as model biomass, for extraction of the valuable compounds from sesame, wheat germ, palm, olive and soybean oils. As a result, the extraction of three free fatty acids (palmitic, oleic and linoleic), α-tocopherol and three phytosterols (campesterol, stigmasterol and β-sitosterol) from six vegetable oils have been demonstrated. The comparison between the published data on the amount of the analysed compounds in the corresponding vegetable oils and presented here extraction results confirm that the developed method allowed not only quantitatively extract the physiologically-active components from various vegetable oils without any pre-treatment but also was reproducible, cost-effective and ecologically-conscious as consumed smaller volumes of organic solvents than commonly used protocols.

Quantitative structure–activity relationship between antioxidant capacity of phenolic compounds and the plasmonic properties of silver nanoparticles

Plasmonic response of silver nanoparticles (AgNPs) reduced with phenolic compounds were evaluated and correlated with the antioxidant capacities of corresponding phenolic compounds and their relative chemical structures. The reference methods including DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging assays were used for the measurement of antioxidant capacity of phenolic compounds which was positively correlated with redox characteristics of these compounds against the formation of AgNPs. It is found that the higher plasmonic response of AgNPs corresponds to the highest antioxidant capacity of phenolic acids, which in turn depends on chemical structures and degree of hydroxylation. The higher degree of hydroxylation in chemical structures of phenolic compounds demonstrated the higher radical scavenging capacity and higher tendency to reduce Ag+ to AgNPs. The influence of reaction time and temperature on reducing efficiency of the tested phenolic compounds is found to be different. Some phenolic compounds such as quercetin, rutin and gallic acid reacted fast (< 1.0 min) while others were found slow reacting. This study establishes the relationship between the antioxidant capacity of phenolic acids and corresponding optical response by means of plamonics, which can be used as an innovative antioxidant detection assay for samples rich in phenolic compounds.

Development and optimization of simplified method of fast sequential HR-CS-FAAS analysis of apple juices on the content of Ca, Fe, K, Mg, Mn and Na with the aid of response surface methodology

A new method for fast sequential measurements of concentrations of Ca, Fe, K, Mg, Mn and Na in apple juices by high resolution-continuum source-flame atomic absorption spectrometry (HR-CS-FAAS) was developed using response surface methodology (RSM). By combination of Box-Behnken response surface design and desirability functions, compromise conditions of simplified sample preparation of apple juices, including 2-fold dilution with water and acidification with HNO3 to 0.8 mol L–1, were established and verified that provided reliable results. The method was applied for analysis of 20 different samples of commercially sold apple juices. Precision (as relative standard deviation for 3 parallel samples, RSD) and accuracy (as relative error in relation to reference values obtained with prior wet digestion of samples) were very good, i.e. from 0.5% to 6% and from –5 to + 6%, respectively.

Robust multi objective optimization of anisotropic yield function coefficients

The coefficients of yield functions have been conventionally determined based on limited number of experiments such as yield stresses and/or R-values along different material orientations. In the present study, a multi objective genetic algorithm (MultiGA) based approach was implemented to obtain coefficients of anisotropic yield functions by simultaneous error minimization for yield stresses and R-values. Three frequently employed yield functions, Hill 1948, Barlat 1989 and Barlat Yld2000-2d, in the sheet metal forming simulations were considered. The performance of the determined coefficients for each yield function was judged by comparing the predicted yield stress and R-values with experimental values. Fundamental questions regarding the effect of experimental data on determining the coefficients were analyzed using the proposed approach. It is generally perceived that increase in experimental data enhances the accuracy of the determination of coefficients. Some counter intuitive results obtained on this regard are discussed. Finally, finite element (FE) simulations were performed to predict earing profile in deep drawn cups and springback profile for split-ring test, which were validated with experimental results. From the comparative study with conventional method and experiment, the yield function coefficients optimized by the MultiGA performed well for predicting the deformation behaviors of various anisotropic sheet metals.

Biofilm architecture on different substrates of an Oculatella subterranea (Cyanobacteria) strain isolated from Pompeii archaeological site (Italy)

Abstract

The Cyanobacterium Oculatella subterranea Zammit, Billi, Albertano inhabits hypogea and stone caves and is a pioneer of different stone substrata. In this study, a strain isolated from the House of Marco Castricio (Archaeological Park of Pompeii, Italy) was identified by a polyphasic approach and used for an in vitro colonization test to verify the influence of the substrate on the biofilm architecture. Fine structure of O. subterranea microbial mats was revealed as well as filaments orientation toward light source. This aim has been achieved through confocal laser scanner microscope microscopy and computer image analysis. Moreover, bioreceptivity of five different substrates, commonly retrieved in archaeological sites of Campania, was assessed for O. subterranea. Our results show that the three-dimensional structure of O. subterranea microbial mats is poorly affected by physical and geochemical features of substrates: in fact, the porous architecture of its biofilm was preserved, independently of the materials. On the other hand, the area/perimeter ratio relative to the O. subterranea growth on tuff, brick, and porphyry showed significant differences, indicating dissimilar levels of bioreceptivity of the three substrates.
