Statistical evaluation of the geochemical variability in overbank sediments in Spain

Abstract

The overall objective of this study is to estimate, detect and specify the main sources of variance which affect the contents of the different elements in overbank sediments across Spain. These sources of variance were assessed and compared by means of a series of analyses of variance (ANOVAs), by regarding two parameters: their significance and their contribution to the total variance. Overbank sediments, sampled in erosion banks, were studied in several locations, in basins which drain different types of geological backgrounds and land uses (urban, mining, agricultural or pristine) across the Iberian Peninsula. Forty-eight elements (mostly in the < 63 μm fraction) were analysed by ICP-OES, ICP-MS and INAA. After an isometric log ratio (ilr) transformation of the data, three ANOVA analyses were performed considering three perspectives: (1) local scale, (2) regional scale: within-profile perspective and (3) regional scale: inter-profile perspective. On a local scale, it was observed that the variability of rare earth elements (REE) depends mostly on the grain size and that heavy metals are also influenced by depth. In the analysis carried out on a regional scale, from a within-profile perspective, depth and duplicates do not influence significantly the variability of the element contents. Finally, from an inter-profile perspective, the selected sources of variance were land use and provenance, whose significance is the highest. While grain size and the selection of depth are of crucial importance in the final results, on local studies, land use and provenance are the ones that influence the most the composition of sediments in regional studies.

Genotype–phenotype correlation, gonadal malignancy risk, gender preference, and testosterone/dihydrotestosterone ratio in steroid 5-alpha-reductase type 2 deficiency: a multicenter study from Turkey

Abstract

Background

Studies regarding genetic and clinical characteristics, gender preference, and gonadal malignancy rates for steroid 5-alpha-reductase type 2 deficiency (5α-RD2) are limited and they were conducted on small number of patients.

Objective

To present genotype–phenotype correlation, gonadal malignancy risk, gender preference, and diagnostic sensitivity of serum testosterone/dihydrotestosterone (T/DHT) ratio in patients with 5α-RD2.

Materials and methods

Patients with variations in the SRD5A2 gene were included in the study. Demographic characteristics, phenotype, gender assignment, hormonal tests, molecular genetic data, and presence of gonadal malignancy were evaluated.

Results

A total of 85 patients were included in the study. Abnormality of the external genitalia was the most dominant phenotype (92.9%). Gender assignment was male in 58.8% and female in 29.4% of the patients, while it was uncertain for 11.8%. Fourteen patients underwent bilateral gonadectomy, and no gonadal malignancy was detected. The most frequent pathogenic variants were p.Ala65Pro (30.6%), p.Leu55Gln (16.5%), and p.Gly196Ser (15.3%). The p.Ala65Pro and p.Leu55Gln showed more undervirilization than the p.Gly196Ser. The diagnostic sensitivity of stimulated T/DHT ratio was higher than baseline serum T/DHT ratio, even in pubertal patients. The cut-off values yielding the best sensitivity for stimulated T/DHT ratio were ≥ 8.5 for minipuberty, ≥ 10 for prepuberty, and ≥ 17 for puberty.

Conclusion

There is no significant genotype–phenotype correlation in 5α-RD2. Gonadal malignancy risk seems to be low. If genetic analysis is not available at the time of diagnosis, stimulated T/DHT ratio can be useful, especially if different cut-off values are utilized in accordance with the pubertal status.

Impaired bone matrix glycoprotein pattern is associated with increased cardio-metabolic risk profile in patients with type 2 diabetes mellitus

Abstract

Purpose

Osteopontin (OPN), osteoprotegerin (OPG) and osteocalcin (OC) are matrix glycoproteins which mediate bone mineralization; moreover, their effects on glucose/insulin homeostasis have recently been demonstrated. Higher circulating OPN and OPG levels have been associated with the presence of insulin resistance, atherosclerosis and coronary heart disease. No data are available on contextual changes of these markers in type 2 diabetes mellitus (T2DM). Therefore, aims of this study were to evaluate serum OPN, OPG and OC levels in T2DM patients and their clinical correlates.

Methods

We recruited 83 consecutive T2DM patients referring to our diabetes outpatient clinics at Sapienza, University of Rome, and 71 non-diabetic sex and age-comparable subjects as a control group. Study population underwent metabolic characterization and carotid ultrasound for intima–media thickness measurement. Plasma OPN, OPG and OC were measured by MILLIPLEX Multiplex Assays Luminex.

Results

T2DM patients had significantly higher circulating OPN and OPG levels than controls (14.3 ± 13.6 vs 10.6 ± 13.7 ng/ml p < 0.001, 0.70 ± 0.60 vs 0.54 ± 4.1 ng/ml, p = 0.02) while OC levels were similar in the two cohorts (6.35 ± 5.8 vs 7.80 ± 7.0 ng/ml, p = n.s). OPN and OPG positively correlated with greater systolic blood pressure (SBP) values, HOMA-IR and HOMA-β, and with the presence of dyslipidemia and carotid atherosclerosis. The association between greater OPN and OPG levels and SBP was independent from possible confounders (both p = 0.01).

Conclusions

Circulating OPN and OPG levels are increased in T2DM patients and identify a particularly unfavourable metabolic profile, mostly expressed by higher SBP. Bone peptides may represent novel markers of vascular stress and accelerated atherosclerosis in diabetes, constituting a possible tool for cardiovascular risk stratification in diabetes.

Fluorene oxidation by solar-driven photo-Fenton process: toward mild pH conditions

Abstract

Polycyclic aromatic hydrocarbons (PAHs) are on the list of priority pollutants to be eliminated from the environment due to their carcinogenic and mutagenic action, chemical stability, and resistance to biodegradation. The aim of this study was to evaluate the degradation of fluorene, a well-known PAH, in aqueous solutions (0.03 and 0.08 mg L−1), by means of a solar-driven conventional (PF) and modified photo-Fenton mediated by ferrioxalate complexes (PFF). Photolysis was also employed for comparison purposes. PF reaction was evaluated at different pH values (2.8, 3.5, and 4.0) and iron concentrations (2, 5, 10, and 20 mg L−1). On the other hand, PFF studies were conducted at mild pH conditions (4.0, 5.0, and 6.0) and iron content of 2 mg L−1, keeping initial iron/oxalate molar ratio at 1:3. In both PF and PFF, the initial hydrogen peroxide/iron molar ratio was maintained at 5. In the presence of methanol as cosolvent for fluorene dissolution, the PF reaction was hampered and no consumption of H2O2 was observed during the reaction carried out at constant pH (2.8). This led to low degradation rates, similar to those achieved by photolysis. Under the same pH but using acetonitrile as cosolvent for fluorene dissolution, fluorene degradation was found to be proportional to the iron content used in the PF experiments. On the other hand, at an invariable iron concentration of 5 mg Fe2+ L−1, the increase in pH was accompanied by a decrease in the molar fraction of the most photoactive iron complex (FeOH2+) and ferric hydroxides precipitation, leading to a reduction in the fluorene degradation rate. With regard to the PFF tests, similar fluorene degradation performance was achieved at pH 4 and 5, while at pH 6 iron precipitation became relevant and the degradation rate was slightly slower. PFF has shown to be more efficient than the PF under the same pH (4) and iron concentration (2 mg L−1). Moreover, even at near neutral pH (6), fluorine degradation was shown to be feasible by using ferrioxalate complexes.
