MicroRNA-183 and microRNA-96 are associated with autoimmune responses by regulating T cell activation

Abstract

MircoRNAs (miRs) are small molecules that regulate gene expression at the posttranscriptional level. They have been proposed to be involved in the regulation of several immune responses including autoimmunity. Here, we identified miR-183 and miR-96 to be highly expressed in CD4⁺ T cells from peripheral blood of Graves' orbitopathy (GO) patients as well as in human and murine T cells upon activation in vitro. By using Luciferase-based binding assays, we identified EGR-1 as target for miR-183 and miR-96. Overexpression of miR-183 and miR-96 in murine CD4⁺ T cells by retroviral gene transfer resulted in decreased EGR-1 and PTEN expression, elevated Akt phosphorylation and enhanced proliferation. In contrast, treatment of murine CD4⁺ T cells with specific antagomiRs increased EGR-1 and PTEN expression and interfered with the proliferative activity upon stimulation in vitro. Strikingly, adoptive transfer of miR-183 and miR-96 overexpressing antigen-specific T cells into INS-HA/Rag2KO mice accelerated the development of autoimmune diabetes, whereas transfer of antagomiR-treated cells delayed the disease onset. These results indicate that miR-183 and miR-96 have the ability to regulate the strength of T cell activation and thereby the development and severity of T cell-dependent autoimmune diseases.

Effects of waste lime and Chinese medicinal herbal residue amendments on physical, chemical, and microbial properties during green waste composting

Abstract

Traditional composting is time-consuming and often results in a low-quality product. The objective of this study was to determine the effects of waste lime (WL; at 0, 2.5, and 3.5%) and/or Chinese medicinal herbal residues (CMHRs; at 0, 10, and 20%) as amendments on the two-stage composting of green waste (GW). The combination of WL and CMHRs improved compost particle-size distribution and pH, decreased nitrogen loss, and increased cation exchange capacity (CEC), nutrient content, and microbial numbers. The combination of WL and CMHRs also accelerated organic matter humification and lignocellulose degradation and therefore increased the germination index of the final compost. Relative to the non-amended compost, the optimal amendment (2.5% WL and 20% CMHRs) increased the percentage of particles of ideal size from 23.8 to 66.9%, the pH from 6.69 to 7.17, the CEC from 52 to 169 cmol/kg, the humic acid to fulvic acid ratio from 1.32 to 2.49, the hemicellulose degradation rate from 42 to 87%, and the cellulose degradation rate from 20 to 61%. The treatment with addition of 2.5% WL and 20% CMHRs to GW required only 21 days to generate the highest quality compost product.

Advances of magnetic nanoparticles in environmental application: environmental remediation and (bio)sensors as case studies

Abstract

Nanotechnology is an emerging technique drawing increasing attentions in biomedical, electronic, environmental, and industrial application. Nanoparticles (NPs) possess unique optical, electrical, catalytic, and thermal properties, among which magnetic NPs (MNPs) are one of the most important groups with excellent superparamagnetism property, large surface area, and biocompatibility. In this review, methods for synthesizing and functionalizing MNPs are summarized and linked to their applications in environmental science as either adsorbents or catalysts for removing contaminants from environmental matrices, illustrating stronger reactivity, higher removal capacity, and fast kinetics. Additionally, we also comprehensively discuss the application of MNPs as (bio)sensors to selectively and sensitively detect the presence of environmental contaminants or pathogenic bacteria. This work summarizes the recent progresses of using MNPs as powerful tools in environmental science and engineering, raising their state-of-art application from environmental perspectives and benefiting researchers interested in NPs and environmental studies.

ABSTRACT

Background

Atopic dermatitis (AD) is an inflammatory skin disease with an associated barrier dysfunction and Staphylococcus aureus infection. The mainstay steroid and calcineurin inhibitor therapy shows some adverse effects. 2,4-Dimethoxy-6-methylbenzene-1,3-diol (DMD) is a benzenoid isolated from Antrodia camphorata.

Objective

We investigated the inhibitory effect of DMD on methicillin-resistant S. aureus (MRSA), the chemokine production in stimulated keratinocytes, and the AD-like lesion found in ovalbumin (OVA)-sensitized mice.

Methods

The antimicrobial effect and cutaneous barrier function were evaluated using an in vitro culture model and an in vivo mouse model of AD-like skin.

Results

DMD exhibited a comparative minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against MRSA with nalidixic acid, a conventional antibiotic. The MIC and MBC for DMD was 78.1 and 156.3 μg/ml, respectively. DMD also showed the ability to eliminate the clinical bacteria isolates with resistance to methicillin and vancomycin. The DNA polymerase and gyrase inhibition evoked by DMD for bacterial lethality was proposed. In the activated keratinocytes, DMD stopped the upregulation of chemokines (CCL5 and CCL17) and increased the expression of differentiation proteins (filaggrin, involucrin, and integrin β-1). Topical application of DMD facilely penetrated into the skin, with AD-like skin displaying 2.5-fold greater permeation than healthy skin. The in vivo assessment using the mouse model with OVA sensitization and MRSA inoculation revealed a reduction of transepidermal water loss (TEWL) and bacterial burden by DMD by about 2- and 100-fold, respectively. Differentiation proteins were also restored after topical DMD delivery.

Conclusion

Our data demonstrated an advanced concept of AD treatment by combined barrier repair and bacterial eradication with a sole agent for ameliorating the overall complications.

Papillary Thyroid Microcarcinoma: Reclassification to Non-Invasive Follicular Thyroid Neoplasm with Papillary-Like Nuclear Features (NIFTP): a Retrospective Clinicopathologic Study

Abstract

Papillary thyroid microcarcinoma (PTMC) accounts for nearly 50% of newly diagnosed PTC cases. There is considerable debate in literature about the clinicopathologic features and prognostic significance of PTMC and whether it should be treated as a separate entity. Due to lack of agreement and supportive data, the consensus study group that established the criteria for non-invasive thyroid neoplasm with papillary-like features (NIFTP) kept its size above 1 cm i.e., excluding PTMC from this new group. As a result, to date, some patients diagnosed with PTMC get aggressive treatments such as partial or total thyroidectomy and even radioactive iodine ablation. We retrospectively studied clinicopathologic features and long-term follow-up of 48 cases of papillary thyroid microcarcinoma. Of these, 7 cases (15%) had capsular invasion, 2 cases (4%) had extrathyroidal extension, 1 case (2%) had lymphovascular invasion, 5 cases (25%) had lymph node metastases, no case (0%) had any distant metastases, and 1 case had recurrence after long-term follow-up (mean 13.7 years, range 1–21 years). Upon slide review, 8 cases fulfilled the criteria for NIFTP and were sub-classified under this new category. These 8 cases had no recurrence after long-term follow-up (mean 12.1 years, range 7–19 years). In this study, we confirmed the previous published reports exhibiting indolent nature of PTMC and also suggested that PTMC cases that fulfill all the criteria for NIFTP can be sub-classified under this term in order to avoid unnecessary aggressive treatment.

Selenate tolerance and selenium hyperaccumulation in the monocot giant reed ( Arundo donax ), a biomass crop plant with phytoremediation potential

Abstract

The response of giant reed (Arundo donax L.) to selenium (Se), added as selenate, was studied. The development, stress response, uptake, translocation, and accumulation of Se were documented in three giant reed ecotypes STM (Hungary), BL (USA), and ESP (Spain), representing different climatic zones. Plantlets regenerated from sterile tissue cultures were grown under greenhouse conditions in sand supplemented with 0, 2.5, 5, and 10 mg Se kg⁻¹ added as sodium selenate. Total Se content was measured in different plant parts using hydride generation atomic fluorescence spectroscopy. All plants developed normally in the 0–5.0 mg Se kg⁻¹ concentration range regardless of ecotype, but no growth occurred at 10.0 mg Se kg⁻¹. There were no signs of chlorosis or necrosis, and the photosynthetic machinery was not affected as evidenced by no marked differences in the structure of thylakoid membranes. There was no change in the maximum quantum yield of photosystem II (F_v/F_m ratio) in the three ecotypes under Se stress, except for a significant negative effect in the ESP ecotype in the 5.0 mg Se kg⁻¹ treatment. Glutathione peroxidase (GPx) activity increased as the Se concentration increased in the growth medium. GPx activity was higher in the shoot system than the root system in all Se treatments. All ecotypes showed great capacity of take up, translocate and accumulate selenium in their stem and leaf. Relative Se accumulation is best described as leaf ˃˃ stem ˃ root. The ESP ecotype accumulated 1783 μg g⁻¹ in leaf, followed by BL with 1769 μg g⁻¹, and STM with 1606 μg g⁻¹ in the 5.0 mg Se kg⁻¹ treatment. All ecotypes showed high values of translocation and bioaccumulation factors, particularly the ESP ecotype (10.1 and 689, respectively, at the highest tolerated Se supplementation level). Based on these findings, Arundo donax has been identified as the first monocot hyperaccumulator of selenium, because Se concentration in the leaves of all three ecotypes, and also in the stem of the ESP ecotype, is higher than 0.1% (dry weight basis) under the conditions tested. Tolerance up to 5.0 mg Se kg⁻¹ and the Se hyperaccumulation capacity make giant reed a promising tool for Se phytoremediation.

Familial Cancer Clustering in Patients with Prolactinoma

Abstract

People are at higher risk for malignancy as they get older or have a strong family history of cancer. This study aims to collect family history of cancer in a large cohort of patients with pituitary adenomas (PA) in outpatient clinic from years 2005–2017. Overall, 46.6% of 1062 patients with PA had a family member affected with cancer. Breast cancer in family members was reported in 15.3% of patients with prolactinomas which was significantly higher than in families of patients with non-functioning pituitary adenomas (NFPA) (10.0%) or acromegaly (6.8%) (p = 0.004). Lung cancer in family members was reported in 12.1% of patients with prolactinomas, significantly higher than in families of NFPA patients (7.0%, p = 0.049). Colorectal cancer in relatives of patients with PA was reported with any type of PA. Furthermore, patients with a positive family history of malignancy were diagnosed with PA at an earlier age than patients with a negative family history (43.6 ± 15.9 vs 46.0 ± 16.4 years, p = 0.015). Female patients with prolactinoma are more commonly diagnosed before the age of 25 years. Forty-two percent of patients with PA diagnosed before the age of 25 years had a second- and third-degree relative with cancer, significantly higher than patients with PA diagnosed later in life (25.8%, p < 0.001). Breast, lung, and colon cancers in second- and third-degree relatives were reported in significantly higher proportion of patients with PA diagnosed before the age of 25 years, compared with patients with PA diagnosed later in life (breast cancer: 10.9 vs 6.1%, p = 0.033; lung cancer: 10.9 vs 5.8%, p = 0.02; colon cancer: 9.5 vs 4.0%, p = 0.004). These results suggest familial cancer clustering in patients with prolactinoma and young patients with PA (younger than 25 years at diagnosis of PA). In particular, there is a strong association between prolactinoma and family history of breast and lung cancers. Further research of possible shared genetic susceptibility of prolactinoma and breast and lung cancers is needed.

Performance of a kerb side inlet to irrigate street trees and to improve road runoff water quality: a comparison of four media types

Abstract

The TREENET inlet is an emerging water-sensitive urban design technology that consists of a novel kerb side inlet coupled with a leaky well infiltration system. The inlets have been retrofitted to existing roads since 2006; however, there is currently little information available on the effectiveness of these inlet and leaky well systems. This study investigated the performance of the kerb side inlets and leaky well system for water quality improvement prior to infiltration to native soil. The leaky wells included four filter media types, namely gravel, water treatment solids, sandy loam and clay. To compare the performance of the four filter media types, batch and column studies were performed in the laboratory. The best performance was observed using the sandy loam as a filter media, followed by clay, water treatment solids and then gravel.
