Functional expression of N-acetyltransferase 1 (NAT1) in differentiated human skin keratinocytes

Abstract

Allergic contact dermatitis to hair dye related molecules represents a considerable health concern. Para-phenylenediamine (PPD) is considered as key sensitizer in hair dye related allergic contact dermatitis. Mechanistically, oxidation of PPD was found to be of great importance for the induction of sensitization¹. Moreover, PPD can be N-acetylated by N-acetyltransferase 1 (NAT1) to mono- and diacetylated PPD in skin^2,3, while other conjugates were rarely detected⁴. N-acetylated PPD metabolites were classified as non-sensitizers in the murine local lymph node assay¹.
